摘  要:目的建立单侧输尿管梗阻(UUO)大鼠肾小管上皮细胞转分化(EMT)模型,检测EMT过程中热休克蛋白47(HSP47)的表达变化。方法采用UUO模型,将雄性SD大鼠36只随机分为假手术组(Sham组)和模型组(UUO组),每组各18只。术后3、7和14 d分别处死两组大鼠。采用HE和MASSON染色法观察各组大鼠术后不同时间肾脏病理改变,用免疫组织化学检测HSP47、波形蛋白(Vimentin)及紧密连接蛋白(ZO-1)的表达。结果随着梗阻时间延长,UUO组大鼠肾小管损伤程度和肾间质纤维化程度逐渐增加,HSP47和Vimentin表达上升(P<0.05),而ZO-1表达下降(P<0.05)。结论 UUO模型大鼠EMT过程中伴有HSP47的高表达,HSP47表达上升可促进EMT过程。[Objectives] To detect the expression of HSP47 in the kidneys of rats with unilateral ureteral obstruction. [Methods] Male Sprague-Dawley (SD) rats were randomly divided into sham group (n = 18) and unilateral ureteral obstructive (UUO) model group (n= 18). The obstructed kidneys were harvested 3, 7 and 14d after UUO. Changes in renal morphology were examined by HE and Masson staining. The expressions of HSP47, vimentin and zona occludens-1 (ZO-1) were examined by immunohistochemistry (IHC) in each group. [Results] Compared with the sham group, the expressions of HSP47 and vimentin were markedly upregulated in the UUO model group (P〈 0.05), but the expression of ZO-1 was downregulated (P〈 0.05), and more severe tubular injury and tubulointerstitial fibrosis were also noticed in the UUO model rats (P〈 0.05). [Conclusions] HSP47 is upregulated in the progress of epithelial to mesenchymal transition (EMT) in the UUO model rats. HSP47 may accelerate this process.
