摘  要:目的研究fat-1转基因对小鼠饮食诱导肥胖的抑制作用及其机制。方法将小鼠分为高脂饮食fat-1转基因和非转基因组及正常饮食fat-1转基因和非转基因组4组。每周记录小鼠体质量和体长,实验结束时称量小鼠内脏周围脂肪质量,测定血清中血脂含量和胃组织内nesfatin-1、ghrelin mRNA的相对表达量。结果转基因小鼠的内脏周围脂肪质量均低于非转基因小鼠(F=20.56~41.75,P〈0.01)。转基因小鼠空腹血糖(FBG)及血清三酰甘油(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)水平均低于非转基因小鼠(F=15.59~94.07,P〈0.01)。转基因小鼠胃组织中nesfatin-1的表达降低(F=18.50,P〈0.01),ghrelin的表达升高(F=14.09,P〈0.01)。nesfatin-1表达与Lee’s指数呈正相关(r=0.621,P〈0.01),ghrelin表达与Lee’s指数呈负相关(r=-0.603,P〈0.01)。结论 fat-1转基因小鼠通过增加内源性n-3多不饱和脂肪酸降低体内n-6/n-3比值,从而调节血脂及肥胖因子的表达,改善机体能量代谢,抑制肥胖的发生。Objective To study the inhibition and its mechanism of fat-1transgenes on diet-induced obesity in mice.Methods The mice in this study were equally randomized to four groups as:transgenic group with high-fat diet and non-transgenic group with high-fat diet;transgenic group with normal diet and non-transgenic group with normal diet.The body mass and stem length of the mice were recorded weekly,on completion of the experiment,the mass of fat surrounding internal organs was measured,and the content of serum lipid and relative expressions of nesfatin-1and ghrelin mRNA in gastric tissue were detected.Results The weight of fat surrounding internal organs in transgenic mice was lower than that in non-transgenic mice(F=20.56-41.75,P0.01).The levels of FBG and serum TG,TC,HDL-C and LDL-C in transgenic mice were lower than that in non-trangenic ones(F=15.59-94.07,P0.01).The expression of nesfatin-1in gastric tissue of transgenic mice declined(F=18.50,P0.01),and that of ghrelin elevated(F=14.09,P0.01).The expression of nesfatin-1was positively correlated with Lee's index(r=0.621,P0.01),and that of ghrelin was negatively correlated with Lee's index(r=-0.603,P0.01). ConclusionAn increase of endogenous n-3polyunsaturated fat acid can reduce the ratio of n-6/n-3in fat-1transgenic mice,regulate serum lipid and the expression of obese factor,improve energy metabolism,and prevent obesity.
