摘 要:目的观察肉毒毒素对膝关节粘连的预防效果及其分子机制。方法将20只Wistar大鼠随机分为实验组和对照组,各10只。实验组给予关节腔内注射A型肉毒毒素(BTX-A),对照组注射生理盐水,术后6周观察两组大鼠膝关节粘连程度,并检测膝关节灌洗液白细胞介素-1(IL-1)、成纤维细胞生长因子(FGF)水平。结果实验组大鼠膝关节粘连程度明显轻于对照组(t=-4.57,P<0.05),膝关节腔灌洗液IL-1、FGF水平显著低于对照组(t=-3.23、-3.59,P<0.05),膝关节腔内粘连组织切片每高倍镜视野成纤维细胞计数显著少于对照组(t=5.95,P<0.05)。结论关节腔内注射肉毒毒素可减轻大鼠膝关节粘连,其机制与抑制IL-1、FGF表达有关。Objective To evaluate the effects and molecular mechanism of botulinum toxin A(BTX-A)in preventing adhesion of knee joint in rats. Methods Twenty Wistar rats were equally randomized to control group and experimental group.BTX-A was injected into the knee joint cavity of rats in the experimental group,and an equal volume of normal saline was injected into that in the control group.The adhesion degree in knee joints of rats in the two groups were observed six weeks after the procedure,and the levels of interleukin 1(IL-1)and fibroblast growth factor(FGF)were detected. Results The extent of knee joint adhesion of rats in the experimental group was milder(t=-4.57,P0.05),the levels of IL-1and FGF in lavage fluid of the knee joints were lower(t=-3.23,-3.59;P0.05),and fibroblast counts of intra-articular adhesion-tissue section of each vision at high magnification were less than that in the control group(t=5.95,P0.05). Conclusion Intra-articular injection of BTX-A can alleviate knee joint adhesion in rats,the mechanism is associated with inhibition of the expressions of interleukin 1and fibroblast growth factors.
