摘  要:目的观察D-柠檬烯对乙醇性肝损伤大鼠抗氧化活性及肝组织核因子κB(NF-κB)、环氧合酶2(COX-2)蛋白表达的影响。方法将雄性Wistar大鼠70只随机分为7组,每组10只。A组为空白对照组;B组为乙醇模型组,每日给予体积分数0.50的乙醇灌胃,前2周8mL/(kg·d),后4周为12mL/(kg·d);C、D、E组分别为D-柠檬烯低、中、高剂量组,乙醇灌胃同时每日分别给予D-柠檬烯100、200、400mg/kg灌胃;F组为水和D-柠檬烯单独高剂量组,每日给予水和D-柠檬烯400mg/kg灌胃;G组为甘利欣对照组,乙醇灌胃同时每日给予甘利欣200mg/kg灌胃。实验进行6周,末次灌胃禁食12h后,取血分离血清,留取肝组织。苏木精-伊红染色观察肝脏组织的病理学改变;检测大鼠血浆和肝匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSHPx)水平;免疫组化法检测肝组织中NF-κB、COX-2表达情况。结果 B组大鼠肝组织呈大泡性脂肪变性,并有大量炎性细胞的浸润;D-柠檬烯各剂量组与B组相比,脂肪变性不同程度减轻,炎性细胞减少。与A组相比,B组血浆SOD、血浆和肝匀浆GSH-Px活力降低,血浆和肝匀浆MDA含量升高;D、E组血浆SOD以及C、D、E组血浆和肝匀浆GSH-Px活力均高于B组,C、D、E组血浆和肝匀浆MDA含量则低于B组(F=42.93~84.37,t=2.73~154.63,P〈0.05)。B组NF-κB、COX-2表达较A组明显增高,而D-柠檬烯各剂量组二者表达均低于B组,差异有统计学意义(F=60.38、49.76,t=9.93~15.25,P〈0.05)。F组大鼠各指标与A组比较差异均无显著性(P〉0.05)。结论 D-柠檬烯能够增强乙醇性肝损伤大鼠体内抗氧化酶的活性,减少脂质过氧化物的产生及肝组织NF-κB、COX-2蛋白表达,从而改善因乙醇暴露引起的肝损伤。Objective To observe the effects of D-limonene on antioxidant ability and the expressions of NF-κB and COX-2in liver of rats with alcohol-induced hepatic injury. Methods Seventy male Wistar rats were evenly randomized to seven groups as A,B,C,D,E,F and G-according to body weight.Group A was the control group;Group B was ethanol-treated group,the rats in this group were given 50%(V/V)ethanol 8mL·kg-1·d-1 for 2weeks following by increasing intake of ethanol up to12mL·kg-1·d-1 for the remaining 4weeks;for groups C,D and E,low,medium and high dose D-limonene(100,200,and400mg/(kg·d))was given,respectively;those in group F were offered water and high dose D-limonene(400mg/kg),intragastrically;those in group G was intragastric administration of diammonium glycyrrhizinate 200mg/(kg·d)same time as ethanol was given,once a day.The experiment lasted for six weeks.After the last intragastric administration,food was prohibited for 12 hours,the blood and liver samples were taken.The pathology of liver tissue was observed by HE staining.the total superoxide dismutase(SOD),malondialdehyde(MDA),and glutathione peroxidase(GSH-Px)were detected.The expressions of NF-κB and COX-2in the liver was detected by immunohistochemical mehod. Results In group B,macrosteatosis and infiltration of a great quantity of inflammatory cells could be seen in the liver tissue;compared with group B,fatty degeneration in group E was lighten,and the number of inflammatory cells decreased.Compared with A group,plasma SOD and plasma and liver homogenate GSH-Px vitality in group B decreased,and the content of MDA in plasma and in liver homogenate increased.Compared with group B,the plasma SOD in groups D and E,and vitality of homogenate GSH-Px in groups C,D and E were higher,and the content of MDA in plasma and liver homogenate was lower(F=42.93-84.37,t=2.73-154.63,P0.05).The expressions of NF-κB and COX-2in group B were higher than that in group A,and in group E,the expressions of both NF-κB and COX-2were lower
