摘  要:目的研究慢性间歇性缺氧(chronic intermittent hypoxia,CIH)对大鼠海马区相关学习记忆能力的影响,并探求其可能的机制。方法 24只SD大鼠随机分为CIH组12只,空白对照组(CON组)12只,CIH组缺氧8周后,2组行Morris水迷宫及穿梭箱实验测试,比较2组大鼠的空间记忆及条件性学习记忆能力;行为检测实验结束后,立刻处死2组大鼠,取脑组织,免疫组织化学方法检测海马区乙酰胆碱M1受体表达水平。结果与CON组比较,CIH组穿越平台次数有所增加,跨越目标象限时间虽升高,但无明显差异(P>0.05)。CIH组第8天回避潜伏期较第2天明显缩短[(362.6±252.4)s vs(546.6±124.7)s,P<0.05]。与CON组比较,CIH组大鼠海马齿状回乙酰胆碱M1受体灰度值显著增高(0.147±0.009 vs 0.072±0.018,P<0.05)。结论 CIH可能通过提高海马区的乙酰胆碱M1受体水平,保持大鼠空间记忆能力。Objective To study the effect of chronic intermittent hypoxia(CIH) on hippocampusrelated learning and memory of rats and its underlying mechanism.Methods Twenty-four SD rats were randomly divided into CIH group and control group(12 in each group).The animals underwent Morris water maze test and shuttle box avoidance test after 8 weeks of hypoxia to compare their spatial memory and conditional learning and memory.The animals were then killed immediately after the tests.Their brain tissue was removed and stained with immunohistochemical method.Expression of CHRM1 in hippocampal dentate gyrus was detected.Results No significant difference was found in spatial memory between control group and CIH group although the delayed memory was impaired in CIH group.The expression level of CHRM1 in hippocampal dentate gyrus was significantly higher in CIH group than in control group(P〈0.05).Conclusion CIH can preserve the spatial memory of rats by upregulating the expression level of CHRM1 in hippocampus.
