摘 要:目的探讨茵陈五苓散含药血清调节血脂的可能机制并筛选其最佳给药浓度及作用时间。方法将40只雄性SD大鼠随机分为菌陈五苓散低、中、高剂量组,辛伐他汀组及空白组,每组8只。菌陈五苓散低、中、高剂量组分别给予4、8、16 g/(kg·d)菌陈五苓散;辛伐他汀组给予1 mg/(kg·d)辛伐他汀片;空白组给予20 mg/(kg·d)生理盐水,各组大鼠每天灌胃2次,连续3天,制备含药血清。取对数生长期的LO2细胞株经过24 h孵化贴壁后随机分为茵陈五苓散低、中、高剂量组,辛伐他汀组及空白组,每组各18孔,每孔分别加入受试血清10μl。同浓度的18孔再分为3组,每组6孔,分别培养24、48、72 h,实时荧光定量法检测各组细胞的低密度脂蛋白受体(LDL-R)mRNA及3-羟基-3-甲基戊二酸单酰辅酶A(HMG-Co A)还原酶mRNA表达情况。结果与空白组比较,茵陈五苓散各剂量组及辛伐他汀组各时间点LDL-R mRNA表达均明显升高,HMG-Co A还原酶mRNA表达均明显下降(P<0.05或P<0.01),其中茵陈五苓散在中浓度、干预48 h的条件下LDL-R mRNA表达达高峰、HMG-Co A还原酶mRNA表达最低。结论茵陈五苓散含药血清调节血脂的机制可能与其能明显上调LDL-R表达、下调HMG-Co A还原酶表达有关,在药物浓度为中浓度、干预时间为48 h时效果最佳。Objective To probe possible mechanisms of serum with Yinchen Wuling San( 茵陈五 苓散) for regulating blood lipids,and to select the best concentration of the drug and action time. Methods 40 male SD rats were randomly divided into low,medium and high dose of Yinchen Wuling San groups,simvastatin group and blank group,with eight rats in each group. Low,medium and high dose of Yinchen Wuling San groups were respectively given 4 g /( kg·d),8 g /( kg·d) and 16 g /( kg·d) of Yinchen Wuling San. Simvastatin group was given 1 mg /( kg·d) of simvastatin. Blank group was given 20 mg/( kg·d) of saline lavage,2 times a day for three days to prepare medicated serum. LO2 cell lines in the logarithmic phase were incubated adherence for 24 h,then were randomly divided into low,medium and high dose of Yinchen Wuling San groups,simvastatin group and blank group,with 18 holes in each group. 10 μl of tested serum was added in each hole respectively. Each 18 holes with the same concentration were divided again into three groups,with 6 holes in each group,and cultivated for 24 h,48 h,and 72 h respectively. Real-time fluorescence quantitative method was used to detect expression levels of the low density lipoprotein receptor( LDL-R) mRNA and 3-hydroxy-3-methylglutary-Co A( HMG Co A) reductase mRNA in cells of each group. Results Compared with the blank group,expression of LDL-R mRNA significantly increased but expression of HMG-Co A reductase mRNA significantly decreased at each time point in each group with different doses of Yinchen Wuling San and the simvastatin group,( P〈0. 05 or P〈0. 01). And the expression of LDL-R mRNA reached the top point but the expression of HMG-Co A reductase mRNA was at the lowest point when medium concentration of Yinchen Wuling San intervened for 48 h. Conclusion Yinchen Wuling San can obviously raise LDL-R expression and lower HMG-Co A reductase expression. This may be its mechanisms of regulating blood lipids.
