摘 要:SGT1参与了细胞周期、逆境反应、蛋白质泛素化和信号转导等过程,在植物抗病反应中起着重要作用.以青花菜(Brassica oleracea var.italica)为材料,克隆到SGT1基因,命名为BoSGT1,其基因组DNA和编码区全长分别为2 007和1 068bp,具有9个内含子;BoSGT1编码了355个氨基酸,编码蛋白含3个TPR、1个CS和1个SGS结构域.BoSGT1与14个SGT1的序列相似性为60%-87%,其中与拟南芥SGT1b的相似性最高;SGT1间的遗传距离为0.003-0.486,BoSGT1与血红老鹳草SGT1、拟南芥SGT1a及SGT1b聚为一组,与其他物种SGT1的关系相对较远,在进化树上处于不同分支.BoSGT1的表达受霜霉菌(Hyaloperonospora parasitica)和核盘菌(Sclerotinia sclerotiorum)的诱导,在霜霉菌侵染下,6,12和24h的表达量增加;在核盘菌诱导下,6,12,24和36h的表达量增加,暗示BoSGT1的表达与霜霉病及菌核病的抗病反应有关.BoSGT1的克隆和表达分析为进一步研究其功能奠定了基础。SGT1is involved in the process of cell cycle,stress response,protein ubiquitination and signaling transduction,and plays an important role in plant disease defense.ASGT1 gene designated as BoSGT1 was isolated from broccoli.The genomic and coding sequences of BoSGT1 were 2 007 and 1 068 bp in length,with nine introns.BoSGT1 encoded 355amino acids containing conserved domains of 3TPRs,one CS as well as one SGS.The identities of BoSGT1 and 14SGT1sof other plants ranged from 60% to 87%,and the highest identity was observed between BoSGT1 and SGT1bfromArabidopsis thaliana.The genetic distance among SGT1 svaried from 0.003 to 0.486.BoSGT1,Geranium sanguineum SGT1,Arabidopsis thaliana SGT1 band SGT1awere grouped together,while other SGT1 swere located at different clades,indicating their relative distinct relationships with BoSGT1.The expression of BoSGT1 was induced by both Hyaloperonospora parasitica and Sclerotinia sclerotiorum.The enhanced expression of BoSGT1 was detected after challenged by Hyaloperonospora parasitica at 6,12 and 24h,and the similar pattern was detected when incubated by Sclerotinia sclerotiorumat 6,12,24 and 36h,revealing its possible function in downy mildew and sclerotinia disease resistance.Cloning and expression analysis of BoSGT1 provide evidences for further studies of its functions.
