摘 要:目的建立高效液相色谱法测定患者血浆中异烟肼和乙酰异烟肼质量浓度的方法,并对目前应用较多的老年肺结核患者进行药代动力学研究。方法 10名老年患者,年龄大于65岁,诊断为肺结核,口服给予异烟肼0.3g,不同时间采集患者血样,用HPLC法测定血浆药物质量浓度,用DAS3.1软件计算药代动力学参数。结果用非房室模型计算出了异烟肼在患者体内的药代动力学参数。结论该方法简单、灵敏、准确,适用于体内异烟肼药代动力学分析。异烟肼在老年患者的药代动力学特征与年轻人群的药代动力学特征存在差异。Objective To determine the concentration of isoniazid(INH)and acetylisoniazid in patient plasma by HPLC and investigate INH′s pharmacokinetics in elder patients.Methods A single 0.3g dose of INH was given to 10 tuberculosis patients with the age more than 65 years old by oral administration route.Blood samples were preceded by protein precipitation and the plasma concentrations of INH were determined by an HPLC method,which was built in this study.Data were analyzed by the DAS3.1software.Results The major pharmacokinetics parameters of INH were calculated by using non compartmental model.Conclusion The method described in this report was of high sensitivity and selectivity for accurate determination of the plasma concentration of INH in human.The results obtained from this study showed that the pharmacokinetics of INH in elder patients was significantly different from that in adult patients.
