摘 要:利用盐溶蛋白PAGE法和SSR分子标记法对玉米杂交种苏玉20和成单60的指纹图谱进行分析。结果发现,应用盐溶蛋白PAGE法可用于苏玉20纯度鉴定,而在成单60的图谱中未出现明显特征谱带,故不能进行鉴定;SSR分子标记法选用了20对SSR引物,分别在苏玉20及其双亲间得到8对引物显示稳定的多态性,而在成单60及其双亲间得到3对,杂种均表现为父母本互补的带型,可作为纯度鉴定候选引物。综合比较,盐溶蛋白PAGE法简单快速,适合单品种检测,而SSR分子标记法适用范围更广,且准确、高效、经济。Salt-soluble protein PAGE and SSR molecular markers analysis were used in seed purity identification of two maize hybrids Suyu 20 and Chengdan 60. The results showed that salt-soluble protein PAGE could be used for purity identification of maize hybrids Suyu 20, not for Chengdan 60, because no distinct salt-soluble protein bands were detected in Chengdan 60; two maize hybrids were subjected to analysis with 20 SSR primers respectively. As a result, eight pairs of primers which could detect polymorphism between Suyu 20 and its parents were determined as candidates for purity identification, and three pairs of primers in Chengdan 60. The bands of hybrid maize were complement types of their parents. In conclusion, salt-soluble protein PAGE had the advantages of simple and quick and applied to a single maize varieties, but SSR technique was more suitable, accurate, efficient and economical.
