摘  要:目的从牛蒡子药材中提取、分离、纯化牛蒡子苷。方法采用正交试验方法优化牛蒡子苷的醇提工艺;使用大孔吸附树脂柱粗分离,经硅胶H层析柱精制,得到牛蒡子苷纯品。结果牛蒡子苷的最佳醇提工艺为乙醇体积分数90%,回流次数3次,回流时间2 h,加8倍溶剂量;以纯水作为大孔吸附树脂柱吸附原液溶剂,收集50%乙醇洗脱液,再经硅胶H层析柱,氯仿-甲醇(9∶1)洗脱,薄层跟踪,收集单点流分,重结晶,得牛蒡子苷纯品,纯度98.23%。结论此方法得到的牛蒡子苷纯品纯度较高,适用于纯度要求较高的实验室或小规模研究。Objective To extract,separate and purify arctiin from medicinal material Fructus arctii. Methods The orthogonal test was adopted to optimize the alcohol extraction process of arctiin. The rude separation was performed by adopting the macroporous resin column and the purification was conducted by a silica gel H column,and then the pure arctiin was obtained. Results The optimal alcohol extraction process of arctiin was 90% ethanol,refluxing for 3 times and 2 h,adding 8 times the amount of solvent; with pure water as the solvent of macroporous resin adsorption original liquid,the 50% ethanol eluent was collected,then eluted by a silica gel H column and chloroform: methanol( 9 ∶ 1),and tracked by TLC,a single point flow was collected and re- crystallized for getting pure arctiin with a purity of 98. 23%. Conclusion The pure arctiin obtained by this method has higher purity and is suitable for laboratory or small-scale research with higher requirement.
