摘 要:目的探讨唑来膦酸对血管的损伤作用及导致颌骨坏死的可能机制。方法3-(4,5-二甲基噻唑。2)-2,5-二苯基四氮唑溴盐[3-(4,5-Dimethyhhiazol-2-y1)-2,5-diphenyltetrazolium bromide,MTTl法检测加入唑来膦酸后血管内皮细胞增殖活性的变化。最高浓度为5000x10~mol/L,进行3倍梯度稀释为2xlO~mol/L,对照组为0mol/L。通过细胞划痕实验观察唑来膦酸对血管内皮细胞迁移能力的影响。细胞黏附实验检测加入唑来膦酸后血管内皮细胞黏附能力的改变。根据MTT实验结果设计浓度分组为高浓度组(5000×10-7moUL)、中浓度组(555×10-7mol/L)、低浓度组(62×10-7mol/L)。结果MTT结果显示在唑来膦酸浓度为0、2×10-7,7×10-7,21×10-7,62×10-7,185×10-7,555×10-7,1667×10、5000×10-7mol/L时,细胞A值分别对应为0.09±0.03、0.82±0.05、0.71±0.06、0.66±0.03、0.64±0.04、0.47±0.04、0.51±0.05、0.35±0.04和0.34±0.11。划痕实验显示24h后细胞迁移距离逐渐减小,空白对照组(38.7±0.42)mm,低浓度组(35.8±4.17)mm,中浓度组(19.9±0.57)mm(P〈0.001),高浓度组(12.5±3.89)mm(P〈0.05),说明唑来膦酸抑制血管内皮细胞迁移能力。黏附实验结果显示对照组、低、中、高浓度组A值分别为1.14±0.18、0.95±0.13、0.81±0.11(P〈0.01)、0.67±0.19(P〈0.001)。实验组结果与对照组比较,当P值〈0.05时表明有统计学意义。结论唑来膦酸对血管内皮细胞的活性、迁移能力、黏附能力有影响,血管形成的障碍可能是引起二膦酸盐骨坏死的机制之一。Objective To investigate the influence of zoledronic acid on vascular endothelial cells.Methods The influence of zoledronic acid on proliferation,migration and adhesion of vascular endothelial cells were tested with 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT),cell migration assay and cell adhesion assay.The results of each experimental group were compared with the control group and the data statistically analyzed.Results In a concentration of 0-0.5 mmol/L,the absorbance value decreased from 0.09 to 0.34 as the drug concentration increased.Scratch test showed that the change of width of scratch before and after 24 hours in control,low,medium and high concentration groups were (38.7± 0.42),(35.8 ±4.17),(19.9±0.57) mm(P〈0.001),(12.5 ± 3.89) mm(P〈0.05).Adhesion test showed that the absorbance value in control,low,medium and high concentration groups were 1.14 ±0.18,0.95±0.13,0.81 ±0.11 (P〈0.01),0.67 ±0.19(P〈0.001).Comparisons between control and experimental groups were analyzed by ttest and P values〈 0.05 were considered statistically significant.Conclusions Zoledronic acid inhibits the proliferation,migration and adhesion of vascular endothelial cells.
