摘 要:目的检测哮喘小鼠呼吸道组织Rac1的表达水平,分析其与nuocytes相关细胞因子表达谱改变的关系,以探讨Rac1调节免疫应答类型和参与哮喘的可能机制。方法建立小鼠哮喘模型,以肺组织、外周血单个核细胞、肺泡灌洗液等为检材,应用实时荧光定量PCR方法分别检测Rac1、IL-5、IL-13、IL-33的mRNA水平;ELISA法检测血清nuocytes相关细胞因子水平。结果哮喘小鼠各组织IL-5、IL-13、IL-33的mRNA水平明显高于对照小鼠,而Rac1表达水平则明显降低,且相关性分析结果显示,气道Rac1的表达水平与IL-5、IL-13、IL-33的表达水平呈负相关。结论高水平表达nuocytes相关因子的哮喘小鼠,伴有呼吸道组织的Rac1表达水平明显降低的趋势,为进一步研究Rac1的免疫调节作用及其与哮喘发生发展的关系奠定了良好的基础。To explore the type and mechanism of Rac 1 immune response, the change of Rac 1 expression and its relationship with nuocytes-associated cytokines, such as IL-5, IL-13 and IL-33, were evaluated in asthmatic mice. The asthmatic mouse model was established and the lung tissue, peripheral blood mononuclear cells (PBMCs) and bronchoalveolar lavage (BAL) fluid were respectively obtained; the mRNA expression levels of Rac 1, IL-5, IL- l3 and IL-33 were determined by quantitative real-time PCR (qRT-PCR). In addition, ELISA was used to detect the levels of nuocytes-associated cytokines in serum. The results showed that the expression levels of IL-5, IL-13 and IL-33 were increased in asthma mouse, while the Racl level in asthma mouse was significantly lower than that in control group. Taken together, there is a significant negative correlation between the expression levels of nuocytes-associated cytokines and Racl, suggesting the Racl level is significantly decreased in asthma model, accompanying with elevation of nuocytes-related factors.
