摘 要:目的研究3种淋巴细胞亚群的辐射效应及其适应性反应。方法采用125I标记的淋巴细胞膜表面的单克隆抗体与细胞膜结合以及PWM(Pokeweed mitogen,美洲商陆有丝分裂原)激活淋巴细胞的方法测量其放射性的变化,以反映γ射线对淋巴细胞的效应及适应性反应。结果 0.5~8.0 Gyγ射线照射后,膜损伤随剂量增加而加重。B淋巴细胞膜损伤程度大于T淋巴细胞,CD8细胞放射敏感性大于CD4细胞。0.048 Gy低剂量辐射导致CD20和CD4细胞的适应性更好。达到8.0 Gy时,适应性反应消失。PWM激活淋巴细胞的方法反映10 Gyγ射线对B细胞的抑制效应非常显著大于CD4细胞。结论攻击剂量损伤CD20〉CD8〉CD4细胞。低剂量照射的适应性反应CD20〉CD4〉CD8细胞。8.0 Gy时适应性反应消失。10 Gy导致PWM细胞的损伤CD20〉CD4细胞。To evaluate the radiation effects and its adaptive response in three lymphocyte subsets in peripheral blood, monoclonal antibodies (McAb) of antigen on membrane of lymphocytes labeled by 125I were used to bind to the cell membrane and lymphocytes were induced by mitogen PWM. The liquid scintillation counter was used to detect the changes of radioactivity of every lymphocyte subsets in peripheral blood. Results showed that challenge dose (0.5-0.8 Gy γ-rays) could cause damage in antigen on membrane of lymphocytes, and with radiation dosage increasing, the damage become more serious. The radiosensitivity of antigen on membrane of B lymphocytes was higher than that of T lymphocytes, and the radiosensitivity of CD8 cell was higher than that of CD4 cell. Low dose radiation of 0.048 Gy could induce adaptive response. The radiation effect of CD20 cell and CD4 were better than that of CD8 cell. At 8.0 Gy, adaptive response of low dose radiation was disappeared. The method of PWM stimulation illustrated that the inhibition effect of 10 Gy γ-rays on B lymphocyte were obviously more than that on CD4 lymphocyte. The result demonstrated the damage degree of challenge dose on lymphocyte membrane were in order of CD20〉CD8〉CD4 cells, while the adaptive response of low dose radiation were CD20〉CD4〉CD8 cells; at 8.0 Gy, adaptive response of low dose radiation was disappeared; 10 Gy γ-rays could cause worse damage in PWM CD20 lymphocytes rather than in PWM CD4 lymphocytes.
