摘 要:目的本研究试图通过检测大麻素(CB)的两种主要水解酶——N-乙酰基乙醇胺水解酸性酰胺酶(NAAA)、脂肪酸酰胺水解酶(FAAH)以及大麻素1、2型受体(CB-1R、CB-2R)在鼠累积损伤性椎间盘慢性炎症反应过程中的表达来探讨其中大麻素的参与过程及作用机制。方法清洁级Wistar大鼠随即分成实验组与对照组,根据Ulrich制作方法准备实验组,以切皮后即缝合为对照组,在术后第3、7、10天,实验组与对照组每次各取6只,取出相应椎间盘,进行形态学观察,应用Real Time-PCR技术检测FAAH、NAAA、CB-1R、CB-2R的表达水平;利用Western blot方法检测CB-2R蛋白含量。结果与对照组相比,实验10 d组可见刺伤所致纤维环断裂,髓核内出血。在椎间盘累积性损伤7 d时,NAAA和FAAH表达含量分别约为对照组的3倍,并形成高峰;而在损伤10 d后,其表达量呈减少趋势。3、7、10 d组CB-2R蛋白表达与对照组相比明显增高(P<0.01)。结论作为水解酶的底物CB类物质在第7天时候表达含量呈低谷,而随着时间的延长逐渐增高。CB受体表达在损伤后显著增高证明CB类物质慢性炎症反应阶段的调节作用越来越大。Objective To reveal the function of CB during disc chronic inflammation caused by cumulative damage through detecting FAAH 、NAAA、CB-1R and CB-2R.Methods Thirty-six Wistar rats were randomly divided into two groups.Prepare the experimental group according to Jill A.Ulrich's methods.6 rats were teken from each group at the time of the 3rd day,7th day,10 th day postoperation,and removed the corresponding intervertebral disc.Morphology was observed.Morphology was observed.Detect FAAH,NAAA,CB-1R,CB-2R by Real Time-PCR and CB-2R protein content by Western Blot.Results Compared with the control group,the annulus rupture and nucleus bleeding were observed in 10 days experimental group.At the time of 7th day after cumulative damage to the disc,NAAA and FAAH expression levels reached a peak at approximately 3 times a gainst the control group,while which showed a decreasing trend at the 7th day after the injury.CB-2R expression increased significantly at the 3rd day,7th day,10 th day experimental group(P〈 0.01) compared with the control group.Conclusion As a substrate for the hydrolysis enzyme,CB expressed at a bottom level at the 7th day postoperatively,but gradually increased over time.The fact that the expression levels of CB-1R and CB-2R were enhanced significantly shows that the regulatory role of CB is growing up over the time of chronic inflammation.
