摘  要:目的:研究一水草酸钙(COM)晶体对肾小管上皮细胞的毒性作用。方法:以永生化的肾小管上皮HK-2细胞作为研究对象,用不同浓度的COM处理细胞,再以DAPI染色并细胞计数研究COM对HK-2细胞增殖的作用;检测细胞培养上清中的LDH含量,说明COM对HK-2的毒性;应用MTS法绘制HK-2细胞增殖曲线;使用流式细胞术检测COM对HK-2细胞的凋亡和坏死的作用。结果:DAPI染色后并细胞计数后HK-2细胞增殖能力经0.1mmol/L的COM处理后便受到抑制,并与对照组比较,差异有显著统计学意义,并随COM浓度的升高,抑制作用更加显著。1mmol/L的COM可使HK-2上清中的LDH显著升高,并在5mmol/L更加明显。1mmol/L的COM处理HK-2细胞后,细胞增殖曲线显著低于对照组,5mmol/L的COM在处理HK-2细胞的第6天几乎全部死亡。0.1mmol/L的COM可使HK-2细胞凋亡显著增加,1mmol/L的COM可使HK-2细胞坏死显著增加,并随浓度的升高而增加。结论:0.1mmol/L的COM刺激HK-2细胞即可使细胞出现生长抑制,浓度达到1mmol/L后,则可出现细胞死亡。Objective:To investigate the cytotoxic effects of calcium oxalate monohydrate (COM) on renal tu- bular epithelium. Method..HK-2 cells were exposed to different doses of COM. Cell proliferation ability were ex- plored via DAPI staining. Then cell toxic effects were demonstrated by LDH essay using cell supernatants. Cell proliferation curves were performed, and the cell apoptosis and necrosis were analyzed by flowcytometry. Result.. DAPI staining showed 0.1 mmol/L COM could inhibit cell proliferation, and it would grow harder as the doses in- creased. The LDH level of the supernatants was significant higher than the control, and it would even higher when the ceils were cultured with 5 mmol/L COM. The cell proliferation curve of 1 mmol/L COM was much lower than the control, and it turned to zero in six days when the COM concentration was 5 mmol/L. A dose of 0.1 mmol/L COM could induce significant ceil apoptosis, while necrosis needed a dose of 1 mmol/L. Conclusion: A dose of 0.1 mmol/L COM could inhibit HK-2 cells proliferation and induce cell death with a dose of 1 mmol/L.
