云芝糖肽对免疫抑制小鼠脾脏组织Foxp3、PD-1和IL-10 mRNA表达的影响

摘　　要：目的探讨云芝糖肽对环磷酰胺诱导的免疫抑制小鼠脾脏组织免疫负调控分子叉状头转录因子3(Foxp3)、程序性死亡因子1(PD-1)和IL-10mRNA表达的影响。方法 18只BALB/c雄性小鼠随机均分为三组。B组云芝糖肽125mg/kg灌胃,每天1次,连续25d;A组和C组以生理盐水灌胃。在灌胃的第17天和第21天,A、B组腹腔注射环磷酰胺150mg/kg制备免疫抑制模型。第26天处死小鼠,取出脾脏,RT-PCR检测脾组织Foxp3、PD-1和IL-10mRNA表达。结果 A组脾脏组织中Foxp3、PD-1和IL-10mRNA水平均高于B组和C组(P<0.01)。B组Foxp3和IL-10mRNA水平高于C组,而PD-1mRNA水平低于C组(P<0.01)。结论云芝糖肽可能通过下调环磷酰胺诱导的免疫抑制小鼠脾脏组织中免疫负调控分子Foxp3、PD-1和IL-10mRNA表达而发挥免疫增强作用。Objective To investigate the influence of polysaccharopeptide on the mRNA expressions of immune negative molecular forkhead/winged-helix transcription factor box protein 3（Foxp3）,programmed death 1（PD-1）and interleukin-10（IL-10）in the spleen of immunosuppressive mice induced by cyclophosphamide.Methods A total of 18BALB/c mice was equally randomized into three groups.Mice in group B were given polysaccharopeptide 125mg/kg by gastric gavage once a day for 25 days.Mice in groups of A and C were given normal saline as the controls.On the 17 th and 21 st day of gastric gavage,cyclophosphamide 150 mg/kg was injected intraperitoneally to induce immunosuppressive mice models in groups of A and B.Mice were killed on the 26 th day for the measurements of the mRNA expressions of Foxp3,PD-1and IL-10 in the spleen by RT-PCR.Results The mRNA expressions of Foxp3,PD-1and IL-10 in group A were higher than those in groups of B and C（P〈0.01）.Foxp3 and IL-10 mRNA expressions were higher,but PD-1mRNA expression was lower,in group B than those in group C（P〈0.01）.Conclusion Polysaccharopeptide may enhance the immunity by downregulating the expression of immune negative molecular Foxp3,PD-1and IL-10 in the spleen of cyclophosphamide-induced immunosuppressive mice.
