摘 要:目的优选和肝理脾颗粒剂的提取工艺。方法分别制备和肝理脾颗粒剂处方药材水煎煮提取物和乙醇提取物,采用HPLC法测定提取物中芍药苷的含量,并比较提取物HPLC图谱差异;采用正交设计优选提取工艺条件。结果水煎煮和乙醇提取物的主要色谱峰基本一致,芍药苷的含量分别为0.138 mg,/mL和0.140mg/mL,无显著差异;乙醇提取物中部分弱极性成分的相对峰面积明显高于水煎煮提取物(P<0.05);乙醇用量、乙醇浓度和提取时间3个考察因素中,提取时间对芍药苷的提取率具有显著影响(P<0.05)。结论和肝理脾颗粒剂药材提取以乙醇回流法优于水煎煮,优选的提取条件为加7倍量60%乙醇回流提取2次,每次2 h。Objective: To optimize the extracting process of Liver-harmonizing and Spleen-regulating Granule. Methods: The water and ethanol extracts of Liver-harmonizing and Spleen-regulating Granule prepared respectively, the difference of HPLC chromatography were compared by determining the paeoniflorin contents in the extracts with HPLC method. The technique condition were optimized by orthogonal design. Results: The main chromatographical peaks of the water and ethanol extracts were basically consistent and the contents of paeoniflorin were 0. 138 mg/mL and 0. 140 mg/mL respectively, there was no significant difference between them ; the peak area of parts of the puny polar components in ethanol extracts markedly wider than that in the water extracts (P〈0.05) ; in the 3 elements observed, namely, the amounts and concentrations of ethanol as well as extracting time, the extracting time had significant influence on the extracting rate of paeoniflorin (P〈0.05). Conclusion: The better technique of extracting Liver-harmonizing and Spleen-regulating Granule is ethanol reflux method and optimizing conditions are twice reflux extractions with 60 % ethanol 7 times more than the former ovemount.
