景天三七对胃出血大鼠的止、凝血疗效观察及最佳剂量筛选

摘　　要：目的研究景天三七对阿司匹林致胃出血大鼠的止血功效及最佳剂量。方法利用阿司匹林构建动物胃肠道出血模型后,用不同剂量的景天三七给予治疗,其中高剂量组用量为30 g/(kg·d),中剂量组为1 g/(kg·d),低剂量组为0.5 g/(kg·d)。期间检测粪便隐血情况,待某剂量组大鼠粪便隐血全部转为阴性,采血观察其血小板数量、PT、APTT及血管内皮素ET-1,并与治疗前相比较,以探索景天三七对胃出血大鼠的血管壁、血小板和凝血因子作用机制及最佳剂量。结果用不同剂量景天三七治疗后,当中间剂量组粪便隐血结果最先全部转阴时,低剂量组中有3只、高剂量组全部动物粪便隐血结果仍为阳性。与治疗前比较,血小板数量明显增高(P<0.05),以中剂量组增加最为显著(P<0.01);PT、APTT无显著变化(P>0.05);ET-1在低剂量组明显增加(P<0.01),中、高剂量组增加不明显(P>0.05)。结论景天三七可通过提高血小板数量,增强血管壁功能达到止血作用,但对外、内源凝血功能无显著影响。使用中等剂量1 g/(kg·d)为增加血小板的最佳剂量。低剂量0.5 g/(kg·d)景天三七可显著增强血管壁功能,但中、高剂量组对血管壁的作用较为平缓。Objective To study the effect of Jingtian Sanqi on hamostatic function and optimal dose in rats gastric hemorrhage induced by aspirin. Methods Establishing animal models of gastric hemorrhage induced by aspirin, then rats were treated with different doses of Jingtian Sanqi. The dosage in three group are 30g/（ kg·d）, 1g/（ kg·d） and 0.5g/（ kg·d）, respectively. Then the feces occult blood was detected during the treatment. In order to explore the hemostatic function and the optional dose of Jingtian Sanqi in rats, platelet count,PT, APTT and ET-1 were observed, and compared with those before being treated to explore the hemostatic mechanism and the optimal dosage of Jingtian Sanqi. Results When treating rats with gastric hemorrhage induced by aspirin with Jingtian Sanqi, occult blood in the feces of the middle dose group showed negative, Meanwhile, 3 rats in low dose group remained positive and so do all rats in the high dose group. Compared with pretherapeutic, the platelet count has increased significantly（ P〈0.05）. Among the 3 dose groups, middle dose group was better than that of the other groups（ P〈0.01）. However, PT and APTT remained almost the same（ P〈0.05） in all groups. It showed significant increase（P〈0.01） in the ET-1 tested in low dosage rats, but there was no change（ P〉0.05） in middle and high dosage groups. Conclusion By increasing the platelet count, Jingtian Sanqi may achieve the effect of hemostasis. Also, in 3 dose groups,middle dose group is better than the other groups. Moreover, Jingtian Sanqi almost has no influence on PT and APTT, while the drug only took effect in low dosage with ET-1, but there was no significantly curative effect in the other groups.
