摘 要:目的:观察淫羊藿苷(Icariin,ICA)对二氯化钴(Co Cl2)诱导PC12细胞缺氧损伤的作用,并初步探讨其作用机制。方法:利用Co Cl2作用于PC12细胞建立化学性缺氧损伤模型;采用噻唑蓝(MTT)法检测细胞存活率;Hoechst33258和Annexin-V/PI染色检测细胞凋亡;利用荧光探针2’,7’-二氯荧光黄双乙酸盐(DCFH-DA)检测细胞内的活性氧(ROS)水平。结果:Co Cl2处理组PC12细胞存活率显著下降,凋亡率和ROS明显增加,而采用2.5~40μM淫羊藿苷预处理30min,呈浓度依赖性抑制Co Cl2(300μM)诱导损伤引起的PC12细胞存活率的降低,凋亡率以及ROS的升高。结论:淫羊藿苷对缺氧诱导的神经细胞损伤有保护作用,其机制可能与其减少ROS的生成有关。Objective: To investigate the effects of icariin(ICA) on apoptosis induced by cobalt chloride(Co Cl2) in PC12 cells. Methods: Co Cl2, chemical-mimetic agent, was used to establish the hypoxia injury model of PC12 cells. The viability of PC12 cells was measured by MTT assay, the percentage of apoptotic cells was assessed by Hoechst33,258 and flow cytometry, The level of reactive oxygen species(ROS) in PC12 cells was measured by fluorescent probe DCFHDA. Results: Co Cl2 induced a decrease in cell viability and an increase in percentage of apoptotic cells in PC12 cells along with overproduction of ROS. However, pre-incubation of icariin for 30 min prior to Co Cl2 treatment significantly rescued the loss of cell viability induced by Co Cl2, decreased the apoptotic percentage of PC12 cells and overproduction of ROS. Conclusion: Icariin protected PC12 cells against Co Cl2-induced apoptosis, which may be associated with the inhibition of overproduction of ROS induced by Co Cl2.
