摘 要:目的:建立稳定的毛菊苣有效部位的色谱条件,确定样品处理方式,确定样品最优检测波长,梯度程序的摸索,以及有效部位中部分单体的确认。方法:确定含药血清的制备方法、采集条件、处理方法,寻找可能的入血成分或代谢产物;对比正常状态下和高脂模型状态下的含药血清,寻找差异入血成分,探明有效的降脂活性物质。结果:发现大部分单体化合物在254nm的吸收都比272nm的紫外吸收要强,试验中确定检测波长为254nm。对比毛菊苣有效部位色谱图与部分单体色谱图及其各个峰的保留时间,发现在9个结构明确的单体当中(CG1、CG2、CG4、CG5、CG7、CG9、CG10、CG14、CG17),有7个存在于毛菊苣有效部位当中(CG1、CG2、CG4、CG7、CG10、CG14、CG17)。结论:实验不仅建立了正常含药血清和高脂血症大鼠模型,观察两种大鼠含药血清及空白血清状态之间的差异,为探索毛菊苣降脂有效部位提供了线索。Aim The objective of this experiment was mainly to establish stable chromatographic conditions of the effective part of Cichorium glandulosum, determine the processing method and optimal detection wavelengt of sample, exploration of gradient program, confirm the part of monomer in the effective fraction. Methods It has been done to determine serum-containing drug preparation method, acquisition conditions, processing method, and look for possible components in plasma or serum containing metabolites. There existed comparison of normal and hyperlipidemic model state, found differences between the constituents absorbed into blood lipid-lowering active substances, and to be proved effective lipidlowering activity substance.Results It was found that the absorption at 254 nm was better than ultraviolet absorption at 272 nm of most single compounds, so the following test to determine the detection wavelength was 254 nm. Retention time comparison of chromatogram of effective parts and partial monomer and each peak of Cichorium glandulosum, found in the 9 monomer structure(CG 1, CG 2, CG 4, CG 5, CG 7, CG 9, CG10, CG14, CG17), there was 7 existing in the effective part of Cichorium glandulosum(CG 1, CG 2, CG 4, CG 7, CG10, CG14, CG17).Conclusion The experiment not only aopted the normal serum containing, also established hyperlipidemia rat models, and it was to examine the difference between two kinds of state of rat serum containing and blank serum, provide clues for exploring the role of lipid-lowering in Cichorium glandulosum effective substance group.
