枸杞多糖对己烯雌酚暴露成年仓鼠睾丸Caspase-8、Caspase-9及p53凋亡蛋白的影响

摘　　要：66只成年雄性仓鼠随机分为空白对照组(A)、DES组(B)、1 mg/kg LBP组(C)、10 mg/kg LBP组(D)、50mg/kg LBP组(E)及VC+VE组(F)。除对照组外,其余5组均皮下注射1mg/kg己烯雌酚,对照组注射等剂量的橄榄油,连续用药7d,同时C、D、E组分别灌服1、10、50mg/kg的LBP,F组灌服100mg/kg的VC及200IU/kg的VE,A、B组灌服等剂量的生理盐水。于末次给药24h后,乙醚麻醉致死仓鼠,取睾丸进行免疫组化检测,确定Caspase-8、Caspase-9和p53的表达。结果显示,通过免疫组化法检测证实,DES引起成年仓鼠睾丸内生精细胞大量凋亡,显著提高Caspase-9的表达(P<0.01);给予不同剂量的LBP后,Caspase-9及p53的表达呈剂量依赖性降低,以LBP高剂量组(50mg/kg)表达最低(P<0.01),同时LBP高剂量组的Caspase-8阳性表达与对照组相比差异不显著。结果表明,枸杞多糖可以降低己烯雌酚所致成年仓鼠生精细胞中凋亡蛋白Caspase-8、Caspase-9和p53的表达,且以50mg/kg剂量作用最为显著。To study the protective effects of Lycium barbarum polysaccharides（LBP）on apoptosis proteins in testis spermatogenic injuries induced by diethylstilbestrol（DES）in hamsters.66 adult male hamsters were randomly divided into control group（A）,DES group（B）,1 mg/kg LBP group（C）,10mg/kg LBP group（D）,50mg/kg LBP group（E）and VC＋VEgroup（F）.Except the control group,the other 5groups were subcutaneous injected 1 mg/kg DES for 7consecutive days,while the control group was injected equivalent amount of olive.Meanwhile hamsters in C,D and E groups were gavaged 1,10 and 50 mg/kg LBP respectively,and the group of F were gavaged 100mg/kg VC＋200IU/kg VE,the groups of A and B were given equivalent amount of normal saline.After 24 hours of the last treatment,the potency of Caspase-8,Caspase-9and p53 in testicular tissue were tested through immunohistochemistry.The results showed that the DES induced sperm cells apoptosis with dramatically increased expression of Caspase-9（P〈0.01）.After treated with different dosage of LBP,the expression of Caspase-9and p53 decreased in a dose dependent manner with 50mg/kg LBP being the best（P〈0.01）.There were no significant difference compared the high dose of LBP group with the control group referring to the expression of Caspase-8.These results indicated that the LBP could downregulate the expression of Caspase-8,Caspase-9and p53 in the testicle of adult male hamster that treated with DES,and the best dose of LBP was 50mg/kg.
