葛根芩连汤对人口腔黏膜上皮细胞增殖的影响

摘　　要：目的探讨葛根芩连汤浸提液对体外培养的人口腔黏膜上皮细胞增殖活性的影响。方法原代培养人口腔黏膜上皮细胞,取第二代培养细胞,随机分为五个实验组和一个对照组。实验组分别用无血清培基(K-SFM)稀释成终浓度分别为2%、10%、15%、20%、50%五个浓度梯度的葛根芩连汤提取液进行培养。对照组仅用无血清培基继续培养。MTT法测定培养的人口腔黏膜上皮细胞的增殖活性。结果原代培养的人口腔黏膜上皮细胞呈多角形,胞核清晰。5个实验组OD值均高于对照组;15%浓度组OD值高于其它4个实验组;以上差异均有统计学意义(P<0.05)。结论葛根芩连汤在15%浓度下能增强体外培养的人口腔黏膜上皮细胞的增殖活性,,有望用于复发性阿弗他溃疡的局部治疗。Objective To evaluate the effects of Gegen Qinlian decoction on the activity proliferation of human oral mucosa epithelial cells cultured in vitro. Method The primary oral mucosa epithelial cells were cultured. Gegen Qinlian decoction was prepared. The second filial generation cultured cells were randomly divided in to five experi- ment groups and one control group. The second filial generation cells Jn experiment groups were cultured with diluted with serum-free medium （K-SFM） to final concentrations of 2 %, 10%, 15 %, 20 %, 50%. The control group was cultured with K-SFM only. The method of MTT was used to determine the cells＇ generation activity. Results The pri- mary cultured cells＇ shape were polygon and had limpid nucelus. The OD values of the experiment groups wre higher than that of the control group, while concentration of ]5% being higher than other experiment groups. The difference was significance. Conclusion The concentrations of 15% of Gegen Qinlian decoction could promote generation of mueosa epithelial obviously,so it may be a good candidate for Recurrent Aphthous Ulcer.
