摘  要:目的探讨三七总皂苷(PNS)对缺血-再灌注大鼠神经功能及海马区葡萄糖转运蛋白-1(GLUT-1)表达的影响。方法将68只健康雄性Wistar大鼠随机分为假手术组(A组),生理盐水对照组(B组)以及PNS治疗小剂量组(C1组50 mg/kg)、中剂量组(C2组100 mg/kg)、大剂量组(C3组150 mg/kg),C1、C2、C3组和B组再根据干预时间点分为0 h、2 h、4 h、6 h四个亚组,每组4只大鼠。线栓法制备大鼠大脑中动脉缺血-再灌注模型,采用行为学评分法评价大鼠神经功能缺损。C1、C2、C3组及B组在脑缺血2 h再灌注0 h、2 h、4 h、6 h分别向腹腔注射相应剂量的PNS和等体积的生理盐水。采用免疫组化法检测大脑缺血2 h再灌注24 h时缺血侧海马区GLUT-1的表达。运用方差分析、t检验等统计方法对数据结果进行统计分析。结果神经功能评分显示:A组神经功能评分为0分;C组大鼠神经功能评分[(1.49±0.42)分]明显低于B组[(3.26±0.44)分](P<0.01),C1[(2.63±0.44)分]、C2[(2.14±0.54)分]、C3[(1.26±0.33)分]三组间比较,C3组神经功能评分最低。免疫组化结果显示,A组缺血侧海马区GLUT-1表达阳性细胞数(11.25±1.48)明显低于B组(16.19±1.38)和C组(37.19±9.8)(P<0.01);相同干预时间点C1组(21.44±2.87)、C2组(39.19±2.63)、C3组(47.94±3.9)脑组织GLUT-1表达阳性细胞数均明显高于B组(16.19±1.19)(P<0.01),且C3组GLUT-1表达阳性细胞数最高;同一剂量PNS组在0 h、2 h、4 h、6 h进行再灌注各时间点之间,缺血侧海马区GLUT-1表达阳性细胞数均无统计学差异(均P>0.05)。结论 PNS可减轻缺血-再灌注大鼠神经功能缺损程度,可能是通过上调GLUT-1的表达,维持脑能量的供应起到神经保护作用的。Objective To investigate the effects of panax notoginseng saponins on neurological function of rats with ischemia reperfusion and on expression of hippocampus ′s glucose transporter 1 ( GLUT-1).Methods Sixty-eight healthy adult male wistar rats were randomly divided into sham-operate group (A),saline control group(B)and PNS therapy group of small-dose group(C1 =50 mg/kg),dose group(C2 =100 mg/kg),high-dose group(C3 =150 mg/kg),the C1,C2,C3 and B group were redivised into ischemia-reperfusion 0 h,2 h,4 h and 6 h subgroups according to the intervention time points ( n=4 in each group ) . By using the suture method to complete the rat middle cerebral artery occlusion model preparation .The neurological deficit in rats was evaluated by using behavioral score .The corresponding doses of PNS or an equal volume of saline were injected intraperitoneally administered at ischemia for 2 h and reperfusion for 0 h,2 h,4 h and 6 h in the PNS groups and the saline control group .Immunohistochemistry assay was used to detect the expressions of GLUT-1 in the hippocampus of ischemic sides at ischemia-reperfusion. Results Behavioral score:the neurological score of group A was 0.The nerve function of group C defect were significantly less than the B group ( P〈0.01 ) .The group C 3 was the least among the three groups C 1 , C2 and C3 .The results of immunohistochemistry assay ,the number of GLUT-1 positive cells of group A was significantly less than group B and C ( all P〈0.01 ) .The number of GLUT-1 positive cells in brain tissue of rats in the same intervention time point of the group C were significantly more than group B ( all P〈0.01 ) , and group C3 was the most.Among different therapy time groups ,the expression of GLUT-1 were basically the same(P〉0.05).Conclusion PNS can lessen the degree of neurologic impairment in ischemia-reperfusion rats by promoting the expression of GLUT-1,and maintain the brain energy .
