摘 要:目的:研究生长因子在肌腱退行性变病理机制中的作用。方法:本研究采用SD大鼠8周下坡跑过度使用模型造成肱二头肌长头肌腱结节沟段退行性变,采用实时定量PCR法检测运动各阶段肌腱胰岛素样生长因子1(IGF-1)、转化生长因子β1(TGF-β1)、骨形态发生蛋白12(BMP12)、碱性成纤维细胞生长因子(b FGF)的基因表达情况。结果:在过度使用中,肱二头肌长头肌腱结节沟段IGF-1基因表达没有显著性变化;TGF-β1、BMP12、b FGF基因在第4周都出现了显著下降,然后又迅速上升。在第8周TGF-β1、BMP12基因表达呈下降趋势,而b FGF基因表达则继续保持上升趋势。结论:与肌腱断裂修复相比,过度使用中肌腱生长因子基因表达紊乱,有可能导致细胞功能异常,推动肌腱退行性变的进程。Objective The purpose of this study was to investigate the role of growth factors in the process of overuse-induced tendon degeneration. Methods Model of overuse of interarticular ligament of articulation of humerus(ILAH)was established in rats through downhill running,and the growth factors gene expression in various stages of overused ILAH was detected by real-time fluorescent quantitative PCR. Results There was insignificant change in IGF-1 gene expression in the intertubercular sulcus of ILAH. The expressions of TGF-β1,b FGF,and BMP12 decreased significantly after 4 weeks of runnung,and then increased quickly. The expressions of TGF-β1 and BMP12 gene tended to decrease,and the b FGF gene expression continued to increase after 8 weeks of running. Conclusion Compared with the repairing process of tendon rupture,the growth factors gene expression in overused ILAH is disordered,which probably leads to the disfunction of cells and advances the process of tendon degeneration.
