摘　　要：目的探讨丙型肝炎病毒(HCV)调节白细胞介素32(IL-32)表达的分子机制。方法将HCV单个基因的表达质粒(p CMV-core、p CMV-p7、p CMV-E1、p CMV-E2、p CMV-NS2、p CMV-NS3、p CMV-NS4A、p CMV-NS4B、p CMVNS5A、p CMV-NS5B)分别与IL-32基因启动子共转染Huh7.5.1细胞,并测定荧光素酶的活性。采用荧光定量PCR检测IL-32细胞内mRNA的表达情况,酶联免疫吸附试验(ELISA)检测细胞上清中IL-32的含量。结果 HCV核心蛋白(core)能够在Huh7.5.1细胞中上调IL-32基因启动子的活性,上调倍数为4.51倍;而且随着HCV core基因的含量不断增加,其调节作用不断增强。与对照组相比,转染core基因后Huh7.5.1细胞的IL-32 mRNA上调3.86倍,细胞上清中IL-32的含量亦明显升高。结论 HCV在Huh7.5.1细胞通过core基因上调IL-32的合成和分泌。Objective To explore the molecular mechanism of interleukin 32 （IL -32） expression regulated by Hepatitis C vi- rus （HCV）. Methods Huh7.5.1 cells were co - transfected with IL - 32 gene promoter and plasmids carrying individual genes （ pCMV - core, pCMV - p7, pCMV - El, pCMV - E2, pCMV - NS2, pCMV - NS3, pCMV - NS4A, pCMV - NS4B, pCMV - NSSA and pCMV- NSSB） of HCV, and the luciferase activity was measured; mRNA expression of IL -32 was measured by real - time PCR; IL - 32 contents in the supernatant of Huh7.5.1 were measured by enzyme - linked immunosorbent assay （ELISA）. Results HCV core protein can upregulate IL - 32 gene promoter activity （4.51 times）, and the up - regulating effect heightened with the increasing of HCV core gene amounts. Compared with the control group, IL - 32 mRNA expression in Huh7.5.1 cells was up - regulated 3.86 times after transfection with core gene, and the protein expression of IL - 32 in supernatants also obviously increased. Conclusion HCV can up - regulate the synthesis and secretion of IL - 32 via HCV core gene.
