摘 要:目的研究制备以PACA(聚氰基丙烯酸烷基酯)纳米材料为载体的头孢氨苄缓释模型,并测定头孢氨苄缓释模型在人工脑脊液中的释放曲线,建立体外释放模型。方法本实验采用重力下沉法制备头孢氨苄缓释模型,采用高效液相色谱法对人工脑脊液中头孢氨苄进行含量测定,测定头孢氨苄的色谱条件:采用C18柱,流动相为水-甲醇-3.86%乙酸钠-4%乙酸(742∶240∶15∶3,V/V),检测波长为254 nm。结果头孢氨苄缓释模型可持续释放达数周以上,体外释放曲线符合零级释放,求取了释放速率、释放时间。结论 PACA纳米材料可作为缓释管,能够有效实现有效组分在PACA纳米材料中的缓慢释放,可作为药物释放载体,为药物长期稳定释放提供简单、有效、经济、实用的方法。Objective To research the preparation of cefalexin sustained release model with PACA ( poly cyano acrylic acid alkyl ester) nano materials as the carrier, and determine the release curve of cefalexin sustained release model in artificial cerebrospinal fluid, and establish release model in vitro. Methods This experiment adopts the gravity sink method for the preparation of cephalexin sustained release model, using high -performance liquid chromatography(HPLC) to determine cefalexin concentrations in artificial cerebrospinal fluid. The chromatographic condition for determination of cefalexin were Cls column and a mobile phase of water-methanol-3.86% acetic acid-4% sodium acetate (742 : 240 : 15 : 3, V/V), and the detection wavelength was set at 254 nm. Results Cefalexin in the sustained - release model could be sustainable release for at least several weeks. Release curve in vitro was consistent with zero - order release, and the release speed and time were calculated. Conclusion PACA nano materials can be used as slow release pipe, and can effectively realize that the effective components in PACA nano materials release slowly. It can be used as release carriers for drugs, and provide a simple, effective, economical and practical model for the drug sustained release stably in a longterm.
