摘 要:目的考察膳食结构是否对氟伐他汀治疗高胆固醇血症的药效有影响。方法半高脂饲料适应性喂养Wistar大鼠5 d,之后采取全高脂饲料喂养25 d复制大鼠高胆固醇血症模型,同时给予氟伐他汀干预,试验过程中调整不同比例高脂饲料喂养,采用全自动生化仪对多个时间段大鼠血清的总胆固醇(TC)、高密度脂蛋白-胆固醇(HDL-C)、低密度脂蛋白-胆固醇(LDL-C)以及HDL-C/TC进行检测,考察氟伐他汀对高胆固醇血症大鼠模型的作用特点。结果半高脂饲料适应性饲养5 d,全高脂饲料继续喂养25 d,大鼠TC和LDL-C水平显著升高,HDL-C/TC值降低,提示高胆固醇血症大鼠模型复制成功。完全高脂饲料喂养组试验第30天以及改半高脂饲料喂养至试验第36天,血脂检测结果显示,氟伐他汀组与模型组相比,TC、LDL-C以及HDL-C/TC值组间差异无统计学意义(P>0.05)。改基础饲料喂养至试验第42天时,氟伐他汀组血清TC、LDL-C和HDL-C水平较模型组显著降低(P<0.01,P<0.05),HDL-C/TC值较对照组显著降低(P<0.05)。结论氟伐他汀对于低脂饮食结构喂养条件下的高胆固醇血症动物模型,降低血清TC和LDL-C水平效果更为显著。Objective To explore the influence of dietary pattern on fluvastatin in experimental hypercholesterolemia rats. Methods Wistar rats were fed with half high-fat diet (HFD) for 5 days adaptively, then fed with HFD for 25 days and treated with fluvastatin at the same time. Subsequently, rats were fed with different proportion HFD throughout the experiment. Plasma levels of TC, HDL-C, LDL-C and HDL-C/TC were detected by automatic biochemical instrument. Results Plasma levels of TC and LDL-C were increased and those of HDL-C/TC were reduced significantly after feeding with half HFD for 5 days adaptively and HFD for 25 days. Compared with model group, fluvastatin showed no effect on the plasma levels of TC , LDL-C and HDL-C/TC at the 30th day (rats were feeding with HFD) and the 36th day(after adjustment for feeding with half HFD) (P 〉 0.05). When rats were feeding with normal diet by fluvastatin at the 42ed day, plasma levels of TC , LDL-C and HDL-C were decreased significantly than model group(P 〈 0.01, P 〈 0.05), and HDL-C/TC were also decreased than that of control group(P 〈 0.05). Conclusion Fluvastatin has effect on decreasing plasma levels of TC and LDL-C for experimental hypercholesterolemia rats feeding with low-fat diet.
