摘 要:目的采用17β-雌二醇联合去势建立大鼠慢性非细菌性前列腺炎模型,检测膀胱逼尿肌蛋白在慢性前列腺炎大鼠膀胱的表达。方法取2月龄SD雄性大鼠60只随机分为6组,每组10只,即:空白对照组、生理盐水组、假手术组、单纯17β-雌二醇组、单纯去势组、17β-雌二醇联合去势组。HE染色判定大鼠慢性前列腺炎模型建造成功后,采用SP免疫组化法、Western blot检测6组大鼠膀胱组织肌动蛋白(α-actin)、肌球蛋白(MLC 2)及高分子量钙调素结合蛋白(hcad)表达的差异。结果免疫组化结果显示17β-雌二醇联合去势组中α-actin、MLC 2、h-cad蛋白表达增多,Western blot显示α-actin在各组的表达,与空白对照组相比,生理盐水组、假手术组、单纯17β-雌二醇组、单纯去势组联合去势组差异有统计学意义。MLC 2、h-cad在各组的表达与空白对照组相比,生理盐水组、假手术组差异无统计学意义,单纯17β-雌二醇组、单纯去势组、联合去势组差异有统计学意义。结论α-actin、MLC 2、h-cad表达变化可能参与慢性前列腺炎排尿相关症状的发病。Objective To establish rats chronic non-bacterial prostatitis model by castration combined injection of 17 beta-estradiol, and to discuss the relationship between chronic prostatitis and bladder dysfunction by the research of detrusor-related proteins. Methods 60 2-month-old SD rats were randomly taken and divided into 6 groups,10 rats for each, as follows: Blank control group, Saline group, Sham operation group, 17 beta Estradiol (0. 25 mg /ml/ kg) group, Simple Castration group, Combined Castration group. To evaluate the results of rat CP model by he-matoxylin-eosin staining, then expression of α-actin, MLC 2 and h-cad in bladder tissues of 6 groups of rats were analyzed via western-blot and S-P immunohistochemical staining. Results Immunohistochemistry showed that ex-pressions of α-actin, MLC 2 and h-cad in Combined Castration group were higher than other groups. Western blot qualitative study showed that expression of α-actin in combined castration group was higher than other groups. Com-pared with the blank control group, expression of MLC 2 and h-cad was moderately elevated in 17 beta estradiol group, simple castration group, and significantly elevated in combined castration group. Conclusion Altered ex-pression of α-actin, MLC 2 and h-cad is likely involved in the pathogenesis of bladder dysfunction following chronic prostatitis.
