摘  要:目的探讨骨桥蛋白(OPN)在系统性红斑狼疮(SLE)肾损害患者Th17细胞异常中的作用。方法取40例健康者和50例SLE患者(其中,伴肾损害SLE患者25例、无肾损害的SLE患者25例的血清和肝素钠抗凝血,用ELISA法检测血清中OPN和白介素-17(IL-17)的表达水平,用流式细胞术检测外周血中CD3+CD8-IL-17A+T细胞的阳性率,并分析各组间差异,以及各因素间的相互关系。结果1 SLE肾损害患者组的OPN、IL-17水平和CD3+CD8-IL-17A+T细胞阳性率显著高于无肾损害的SLE患者组、健康对照组,差异均有统计学意义(P<0.01);无肾损害的SLE患者组和健康对照组间OPN、IL-17、CD3+CD8-IL-17A+T细胞阳性率的差异均无统计学意义(P>0.05)。2血清中OPN分泌与IL-17水平、CD3+CD8-IL-17A+T细胞阳性率之间均成明显正相关性(P<0.01)。结论外周血中的OPN对SLE肾损害患者的Th17细胞异常起着重要的调控作用。Objective To investigate the regulatory role of osteopontin (OPN) in abnormal Th17 cells of systemic lupus erythematosus (SLE) patients. Methods The levels of OPN and interleukin-17(IL-17) in serum and the rate of CD3 + CD8 - IL-17A + T cell from 40 healthy controls and 50 SLE patients (25 SLE patients with renal dam-age and 25 SLE patients without renal damage) were detected by ELISA and flow cytometry methods. Besides, difference between groups and interaction among multiple elements were analyzed. Results ① The levels of OPN and IL-17 in serum, the rate of CD3 + CD8 - IL-17A + T cell in SLE patients with renal damage all obviously increased than SLE patients without renal damage and healthy controls (P 〈 0. 01), while there was no difference between SLE patients without renal damage and healthy controls (P 〉 0. 05). ② Meanwhile, significant positive correlation was shown between serum OPN and serum IL-17 level, CD3 + CD8 - IL-17A + T cell (P 〈 0. 01). Con-clusion OPN plays an important regulatory role in abnormal Th17 cells of SLE patients with renal damage.
