摘  要:目的研究海南鱼泡草总黄酮(total flavonoids of Jussiaea repens,TFJ)对小鼠慢性皮炎的治疗作用。方法用不同浓度二硝基氯苯(2,4-dinitrochlorobenzene,DNCB)反复刺激昆明小鼠皮肤致敏后,灌胃给药不同剂量的TFJ。再通过H&E染色检测皮肤组织病理学变化,ELISA检测血清总Ig E和IL-2含量变化,RT-PCR检测皮肤细胞因子Eotaxin-1/CCL11和LARC/CCL20的m RNA表达。结果通过TFJ和氢化可的松治疗后,TFJ能减轻鼠耳肿胀度,减少肥大细胞数及浸润炎症细胞数,并且能降低小鼠血清中升高的Ig E水平、升高血清中降低的IL-2水平,同时降低趋化因子配体基因Eotaxin-1/CCL11和LARC/CCL20的m RNA表达。结论 TFJ能有效抑制小鼠慢性皮炎。Jussiaea repens is a traditional Chinese folk medicine with anti-inflammatory properties. Atopic dermatitis(AD) is an inflammatory skin disease associated with enhanced T-helper2(Th2) lymphocyte response to allergens that results in elevated serum eosinophil and immunoglobulin E(Ig E) levels and leukocyte infiltration in atopic skin sites. This study designed to investigate the effect of total flavonoids of Jussiaea repens(TFJ) on 2,4-dinitrochlorobenzene(DNCB) induced AD mouse models, by observing the effects of TFJ on blood eosinophils, skin mast cells, production of serum Ig E, and expression of cytokine m RNA in the atopic dermatitis skin lesions of DNCB-treated mice. Data showed that TFJ plus hydrocortisone significantly reduced blood eosinophils cell numbers in DNCB-treated mice; histological analysis demonstrated that TFJ could decrease mast cell count and dermal infiltration inflammatory cells. In the skin lesions, m RNA expressions of Eotaxin-1/CCL11 and LARC/CCL20 were inhibited by TFJ; TFJ also suppressed the production of serum Ig E level but raised the serum IL-2 level in DNCBinduced atopic dermatitis model. Taken together, our results showed that gastric administration of TFJ benefits the therapeutic effects in AD symptoms, suggesting that TFJ might be a useful candidate for the treatment of AD.
