摘  要:目的寻找AITD患者血清中异常表达的循环microRNA,分析其表达特征。方法本研究选择了Graves病患者41例,桥本甲状腺炎患者36例,正常对照人群41例。利用microRNA芯片的方法筛选各组人群血清中循环表达的microRNA。对表达异常并与免疫功能相关的microRNA,用q RT-PCR的方法进行验证;同时分析这些microRNA与TSH水平的相关性。结果研究发现与正常对照人群相比,分别有17种和16种microRNA在GD患者和HT患者血清中异常表达。其中在2种患病人群中均异常表达,且与人体免疫功能相关的microRNA有4种,分别是mi R-155、mi R-146、mi R-21和mi R-451,q RT-PCR结果验证该mi RNAs在患病组中均高于正常人群。进一步分析发现mi R-155与患者血清TSH水平呈负相关。结论 AITD患者血清中存在异常表达的mi RNAs,mi R-155具有作为反映甲状腺功能状态的新的血清学标志物可能性。To characterize the different circulating mi RNA expression in patients with AITD, we recruited forty-one patients who met criteria for GD, thirty-six patients of HT, and 41 healthy controls. The expression pattern of mi RNAs in serum obtained from GD patients, HT patients and healthy controls were determined by using Microarrays. Four interesting mi RNAs(mi R-155, mi R-146, mi R-21 and mi R-451) were selected and validated by Taq Man-based real-time PCR. And the relationships between the expression level of the above four mi RNAs and the serum TSH level were analyzed by statistical methods. We found seventeen and sixteen mi RNAs that were differently expressed in serum from patient with GD and HT respectively, compared with healthy subjects by microarray analysis. q RT-PCR data consistently confirmed that serum level of mi R-155, mi R-146, mi R-21 and mi R-451 were increased in both GD and HT patients compared with healthy control. And the expression level of mi R-155 was negatively correlated with the serum TSH level. Our results reveal that differential expressions of serum mi RNAs are exist in AITDs patients, and the expression level of mi R-155 is associated with the function of thyroid, which may serve as biomarkers of AITD for diagnostic or treatment.
