摘 要:目的:研究ERK信号通路在脑缺血耐受诱导中的作用,观察疏血通脉胶囊预处理对其调控作用。方法:对大鼠行3 min脑缺血预处理,诱导其产生脑缺血耐受,24 h后建立脑缺血再灌注模型(缺血预处理组),观察ERK/P-ERK的变化情况,并与假手术组、缺血再灌注组、疏血通脉胶囊组及PD98059组进行比较,检测各组神经元凋亡数量,观察ERK/P-ERK与神经元凋亡的相关性。结果:缺血预处理组及疏血通脉胶囊组P-ERK表达水平均明显上调,同时神经元凋亡数量减少,与缺血再灌注组比较差异有统计学意义。腹腔注射ERK抑制剂PD98059后,大鼠P-ERK表达水平受到明显抑制,神经元凋亡数量增加,并加重神经功能缺损情况。结论:脑缺血预处理能够减少脑缺血再灌注后神经元凋亡,改善神经功能,其机制可能与ERK信号通路激活有关,疏血通脉胶囊预处理可能通过激活该通路起到脑保护作用。Objective: To explore the function of ERK( extracellular-signal regulated kinas) signaling pathway in the induction of brain ischemic tolerance, and observe the function of Shuxue Tongmai capsule pretreatment.Methods: Ischemic preconditioning was performed for three minutes on the rats to induce the cerebral ischemic tolerance. Rat model of cerebral ischemia reperfusion( the ischemia pretreatment group,I/R group) was established24 hours later. Western blot was used to detect the protein expression of ERK and P-ERK( phosphorylation extracellular-signal regulated kinas),comparing the expression with the sham operation group,I/R group and Shuxue Tongmai capsule group and PD98059 group. TUNEl method was applied to detect the apoptosis of neurons and we also study the relationship between expression of ERK,P-ERK and the apoptosis of neurons. Results The expression of PERK in ischemia preconditioning group increased significantly and the apoptosis of neurons quantity declined. It was significantly to the I/R group. After given the PD98059,the ERK inhibitor,the expression level of P-ERK was restrain significantly and the apoptosis of neurons deteriorate significantly. Conclusion: Ischemia preconditioning can decrease the apoptosis of neurons in cerebral ischemia reperfusion,and improve neurologic function. Its mechanism related to the activation of the ERK signaling pathway. Shuxue Tongmai capsule pretreatment can protect the cerebral by the ERK signaling pathway.
