摘  要:目的:观察去卵巢骨质疏松症大鼠肾组织Msx2 mRNA及蛋白表达,探讨绝经后骨质疏松症(Postmenopausal Osteoporosis,PMOP)的发病机制以及鹿茸中药复方的疗效机理。方法:去卵巢复制骨质疏松症大鼠模型,实验设正常组、模型组、假手术组、鹿茸中药复方组、钙尔奇D组、骨疏康组,灌胃给药12周。应用双能X线骨密度仪检测骨密度,实时定量RT-PCR及Western Blot检测Msx2 mRNA及蛋白表达。结果:与正常组比较,模型组股骨骨密度明显降低、肾组织Msx2 mRNA及蛋白表达明显降低;与模型组比较,鹿茸中药复方组、骨疏康组股骨骨密度明显升高;鹿茸中药复方组、钙尔奇D组、骨疏康组肾组织Msx2mRNA表达明显上调;鹿茸中药复方组肾组织Msx2蛋白表达明显上调;与钙尔奇D组、骨疏康组比较,鹿茸中药复方组肾组织Msx2 mRNA及蛋白表达均明显升高。结论:PMOP的发病机制之一可能是肾组织Msx2 mRNA及蛋白表达降低,鹿茸中药复方可能通过上调肾组织Msx2 mRNA及蛋白表达,有效防治PMOP,其作用优于钙尔奇D和骨疏康。Objective: To discuss the pathogenesis of postmenopausal osteoporosis( PMOP) and the therapeutic effect and mechanism of pilose antler traditional Chinese medicine compound by Msx2 mRNA and protein expression in kidney tissues of ovariectomized osteoporosis rats. Methods: We established the osteoporosis rat models by ovariotomy. We divided all the rats into normal group,model group,sham-operated group,pilose antler traditional Chinese medicine compound group,GAIERQI D group and GUSHUKANG group. After 12 weeks of administrating by gavage,we used dual energy X-ray absorptiometry to detect bone mineral density,Real-Time Quantitative RT-PCR to detect Msx2 mRNA expression and Western Blot to detect Msx2 protein expression. Results: Compared with normal group,femur bone mineral density of model group decreased significantly,Msx2 mRNA and protein expression decreased significantly in kidney tissues. Compared with model group,femur bone mineral density of pilose antler traditional Chinese medicine compound group and GUSHUKANG group increased significantly; Msx2 mRNA expression in kidney tissues of pilose antler traditional Chinese medicine compound group,GAIERQI D group and GUSHUKANG group up-regulated significantly; Msx2 protein expression in kidney tissues of pilose antler traditional Chinese medicine compound group up-regulated significantly. Compared with GAIERQI D group and GUSHUKANG group,Msx2 mRNA and protein expression in kidney tissues of pilose antler traditional Chinese medicine compound group both increased significantly. Conclusions: Maybe it is one aspect of the pathogenesis of PMOP that Msx2 mRNA and protein expression in kidney tissues decreases; Pilose antler traditional Chinese medicine compound has better effects of up-regulating Msx2 mRNA and protein expression in kidney tissues to prevent and treat PMOP than GAIERQI D and GUSHUKANG.
