摘 要:目的:探讨冬胃颗粒对寒邪客胃型胃溃疡大鼠血清细胞因子水平的影响。方法:采用乙酸注射法制备大鼠胃溃疡模型,采用寒冷因素刺激复制寒邪客胃型动物模型,前3 d先用0℃冰水浸泡,每次10 min,后3 d用0℃冰水灌胃,治疗组同时给予药物灌胃。观察各组大鼠的症状、体征,测定大鼠胃黏膜溃疡指数(UI)及溃疡抑制率,应用ELISA法检测血清细胞因子含量。结果:冬胃颗粒组大鼠表现与大便性状优于西药组。药物治疗可使溃疡缩小,冬胃颗粒组优于雷尼替丁组。与正常对照组比较,溃疡模型及寒性溃疡模型大鼠血清6-keto-PGF1α水平降低,ET-1水平增高;与寒性溃疡组比较,治疗组可明显升高6-keto-PGF1α及EGF水平,降低ET-1水平,中药治疗组6-keto-PGF1α及EGF水平高于西药治疗组,ET-1水平低于西药治疗组。结论:寒性刺激可增加胃溃疡大鼠胃黏膜损害,冬胃颗粒通过调整攻击因子与保护因素平衡起到保护作用。Objective: To study the influence of Dongwei Granule on the levels of cytokines in serum of Acetic Acidinduced Gastric Ulcer in Rats with cold stimulation. Methods: The rat model of gastric ulcer was constructed by injection of acetic acid. The rats were stimulated by cold,filled the stomach with 0℃ ice water 3 days after soaked with 0℃ ice water3 days. The rats of treatment groups were filled the stomach with Chinese or western medicine. The changes of rat superficial signs and symptoms and ulcer sizes were recorded to calculate the ulcer index( UI) and ulcer inhibitory rate.And the levels of EGF,6-keto-PGF1 αand ET-1 were detected by ELISA. Results: The signs and symptoms of treatment groups were better than ulcer model group. The treatment of Ranitidine and Dongwei granule can reduce the ulcer area of rat model. Compared with gastric ulcer model of cold stimulation group, the UI of treatment groups was reduced significantly. The ulcer inhibitory rate of Dongwei granule treatment group was better than Ranitidine treatment group. The levels of 6-keto-PGF1 αof model of gastric ulcer group and gastric ulcer model of cold stimulation group were decreased significantly,and the levels of ET-1 were increased significantly compared with normal control group. The levels of 6-ketoPGF1 αof gastric ulcer model of cold stimulation group were lower than model of gastric ulcer group. Compared with gastric ulcer model of cold stimulation group,the levels of 6-keto-PGF1 αand EGF in serum of treating groups were increased significantly,the levels of ET-1 Were decreased significantly,the levels of 6-keto-PGF1 αand EGF of Dongwei granule treatment group were higher than Ranitidine treatment group,and the levels of ET-1 were lower. Conclusion: Cold stimulation can aggravate the damage of gastric mucosa of gastric ulcer rats,and Dongwei granule can alleviate the damage through re-establishing the balance of attack factors and protective factors.
