摘  要:目的:探究姜黄素在Ang II诱导的心脏成纤维细胞大量增殖和CTGF、PAI-1过表达中的影响,探讨姜黄素与心脏成纤维细胞增殖的关系。方法:心脏成纤维细胞原代的培养,分别用倒置显微镜及免疫组织化学法对原代心脏成纤维细胞进行监测,用MTT法测定不同浓度姜黄素(0-100μmol/L)对心脏成纤维细胞活性的影响,用MTT法检测不同浓度姜黄素对心脏成纤维细胞增殖能力的影响,用real-time PCR检测CTGF、PAI-1mRNA,用western-blot法分析不同浓度姜黄素对各组心脏成纤维细胞CTGF、PAI-1表达的影响。结果:在0-60μmol/L范围内,姜黄素未显示明显的细胞毒性,因此在后续试验中我们选择5、10、30μmol/L浓度姜黄素进行试验,试验说明姜黄素可以有效的抑制Ang II诱导的心脏成纤维细胞增殖,并具有浓度依赖性,可以有效地抑制Ang II诱导的CTGF和PAI-1表达。结论:姜黄素能够有效地抑制Ang II诱导的心脏成纤维细胞过度增殖和CTGF、PAI-1 mRNA与蛋白的过表达,从而起到抗增殖作用。objective: Explore curcumin proliferate and CTGF,PAI-1 overexpression in Ang II-induced effects on cardiac fibroblasts. Explore the relationship between curcumin and the proliferation of cardiac fibroblasts. Method: The primary cultured cardiac fibroblasts,respectively by inverted microscope and immunohistochemical method for primary cardiac fibroblasts were monitored. Determined by MTT method is used to test different concentrations of curcumin( 0 - 100 mu mol/L) effect on the activity of cardiac fibroblasts. The effect of different concentration of Cur( 0 - 100 μmol/L) on the cell proliferation ability of cardiac fibroblasts was measured by MTT assay. The levels of mRNA and protein of CTGF,PAI-1,were measured by real-time PCR and western-blotting. Result: Curcumin exhibits no cellular toxicity on cardiac fibroblasts at the concentration between 0 - 60 μmol/L. In our study,we chose the concentration of 5,10,and 30 μmol /L to treat the cell to observe whether curcumin can inhibit Ang II-induced over-proliferation and overexpression of CTGF and PAI-1 in cardiac fibroblasts. Conclusion: Curcumin inhibits Ang II-induced cardiac fibrosis by suppressing over proliferation and the overexpression of CTGF and PAI-1 in Ang II-stimulated cardiac fibroblasts.
