摘 要:目的:探讨花楸猕猴桃根对肝细胞CYP1A1酶的作用机制。方法:40正常SD大鼠随机分为空白对照组和猕猴桃根提取物高、中、低剂量组(50 g/kg、25 g/kg、13 g/kg),连续灌胃7 d,2次/d,处死大鼠后取出肝脏,通过检测N-脱甲基酶活性反映细胞CYP1A1酶活性,提取组织RNA逆转录后用RT-PCR检测CYP1A1mRNA表达水平。结果:与空白对照组相比,花楸猕猴桃根高、中、低剂量组CYP1A1酶活性均明显受到抑制,且呈浓度依赖性(P<0.05),但各剂量组间无显著性差异;与空白对照组比较,花楸猕猴桃根高、中、低药物剂量组能显著抑制CYP1A1 mRNA的表达(P<0.05)。结论:花楸猕猴桃根可下调大鼠肝细胞CYP1A1酶的活性,并抑制其mRNA表达水平,这可能是花楸猕猴桃根能够预防癌症的作用机制。Objective:To explore the effects of roots of Actinidia sorbifolia C. F. Liang on CYP1A1 in hepatocyte of rats and the mechanism. Methods:The 40 Wistar rats were randomly divided into 4 groups:blank control group,high dose of root group,middle dose of root group and low dose of root group. The rats received intragastric administration of water decoction of roots of Actinidia sorbifolia C. F. Liang( high dose group 50 g/kg,middle dose group 25 g/kg,low dose group 13 g/kg,repective-ly. )continuously for 7 days,twice daily. The rats were killed and liver tissue was taken to detect the enzymatic activity of CYP1A1 by detecting activity of ADM,electrophoresis method was adopted to check the integrity of extracted RNA sample,and RT-PCR was used to detect the mRNA expression level of CYP1A1. Results:The experimental results showed that compared with blank group,the en-zymatic activity of CYP1A1 in high,middle,and low dose group was significantly inhibited in a dosage dependant manner(P﹤0. 05),but there was no significant differences between high,middle and low dose group. Besides,the mRNA expression levels of CYP2A1 were inhibited in high,middle,and low dose group(P﹤0. 05). Conclusion:The roots of Actinidia sorbifolia C. F. Liang can down-regulate the enzymatic activity of CYP1 A1 and inhibit the mRNA expression level of CYP1 A1 ,which is possibly related to its prevention of cancer.
