摘 要:目的观察肿节风注射液对小鼠T细胞淋巴瘤EL-4细胞的抑制效应。方法体外实验中,EL-4细胞经不同浓度肿节风注射液处理48 h后,采用MTT、吖啶橙/碘化丙啶(AO/PI)染色法和流式细胞术(FCM)观察细胞生长抑制及凋亡。体内实验中,分设正常组,模型组,环磷酰胺组(25 mg·kg-1),肿节风注射液低、高剂量组(300,500 mg·kg-1),每组5只,腹腔注射给药,连续10 d,观察小鼠一般状况,测量肿瘤生长,计算抑瘤率。结果 MTT结果表明,肿节风注射液体外对EL-4细胞生长呈浓度依赖性抑制,半抑制浓度IC50值为(36.98±0.97)mg·m L-1。浓度在12.5,25和50 mg·m L-1时EL-4细胞的凋亡率分别为10.3%,31.7%和45.2%。低剂量(300 mg·kg-1)和高剂量(500 mg·kg-1)肿节风组的抑瘤率分别为11.9%和37.1%,与模型组比较,差异均有统计学意义(P<0.05)。肿节风治疗组荷瘤小鼠在一般状况、体质量、血细胞计数等方面未见明显毒副作用。结论肿节风注射液对小鼠T细胞淋巴瘤EL-4细胞具有一定抑制效应,值得进一步深入研究。Objective To observe the inhibitory effects of Zhongjiefeng(Herba Sarcacandrae) injection on EL-4 cell line isolated from mouse T cell lymphoma. Methods In in-vitro assay, EL-4 cell growth inhibition and apoptosis were determined by MTT, acridine orange/propidium iodide AO/PI staining and flow cytometry(FCM) after treatment with Zhongjiefeng injection for 48 h. In in-vivo assay, mice were randomly divided into five groups, namely normal group, model group, cyclophosphamide (25 mg·kg-1) group, and low- and high-dose Zhongjiefeng injection(300, 500 mg·kg-1) groups. Each group had 5 mice, and the medication was by intraperitoneal injection for 10 continuous days. During the administration, mice's general condition was observed daily, tumor growth was observed and tumor inhibition rate was calculated. Results The results of MTF assay showed that the inhibitory effects of Zhongjiefeng injection on EL-4 cell was concentration-dependent, with IC50 being (36.98 ± 0.97) mg·mL-1. The apoptosis rate of EL-4 induced by 6.25, 12.5, 25 and 50 mg·mL-1 of Zhongjiefeng injection was 10.3 %, 31.7 % and 45.2 %, respectively. Compared with the model group, the tumor-growth inhibition rate in low- and high-dose Zhongjiefeng injection groups was 1.2 %, 11.9 % and 37.1% respectively, the difference being statistically significant(P 〈 0.05). No toxic or side effects were shown in Zhongjiefeng injection groups. Conclusion To some extent, Zhongjiefeng injection has inhibitory effects on mouse T cell lymphoma cell line EL-4, and its mechanism need to be studied further.
