摘 要:目的探讨自然铜炮制促进骨折愈合作用及机制。方法建立家兔骨折模型,观察自然铜生品、煅品对血清生化指标、骨密度、骨痂中微量元素及骨组织形态的影响。结果在给药后3周煅品高、低剂量组血清碱性磷酸酶(ALP)活性,血磷(P)、血清钙磷乘积([Ca×P])均显著高于模型组(P<0.05,P<0.01)和生品组(P<0.05,P<0.01);与模型组比较,给药后3周两组煅品骨密度有显著性升高(P<0.05);与模型组比较,给药后3周煅品高剂量组骨痂中Fe和Cu均显著升高(P<0.05)。X线结果显示,与模型组比较,煅品高、低剂量组能明显促进骨痂生长。结论自然铜煅品促进骨折愈合疗效显著优于生品,且主要作用于骨折中期,其作用机制可能是通过促进成骨细胞合成、分泌ALP,增加血磷含量,促进钙盐沉积,增加微量元素的吸收、增强骨密度,从而促进骨折的愈合。Objective To compare the effect of pyritum before and after Rabbit model of radius fracture was established. After oral administration processing on fracture healing. Methods of raw and calcined pyritum, the serum biochemical indexes, bone mineral density (BMD), the microelement in callus, and callus growth in rabbits were observed. Results Compared with model group and raw pyritum group, a significant increase of alkaline phosphatase (ALP), serum phosphorus (P) and serum calcium-phosphorus product [Ca × P] was showed in high-, low-doseprocessed groups 3 weeks after administration (P 〈 0.05, P 〈 0.01). Compared to the model group, BMD in processed pyritum groups was noticeably high 3 weeks after administration(P 〈 0.05). Trace element Fe, Cu contents in the callus were proved to be higher in high-dose of processed group than those in the model group 3 weeks after administration(P 〈 0.05). The results of X-ray photography showed that callus growth in high- and low-dose processed pyritum groups was more obvious than the model group. Conclusion Processed pyritum has better effect of promoting fracture healing than raw pyritum. The possible mechanism is related to promoting the synthesis and secretion of ALP in osteoblasts, enhancing the serum P content, accelerating the deposition of calcium salt, promoting the absorption of trance elements and increasing the BMD in the middle period of fracture healing.
