摘  要:目的考察灯盏生脉胶囊对氯吡格雷及其代谢物SR26334在大鼠体内药动学的影响。方法将SD大鼠随机分为氯吡格雷组和氯吡格雷加灯盏生脉胶囊组,测定给药后不同时间点氯吡格雷及SR26334在血浆中的含量。结果与氯吡格雷组比较,氯吡格雷加灯盏生脉胶囊组血浆中氯吡格雷最大血药浓度(Cmax)降低,平均驻留时间(MRT0-t)延长,差异有统计学意义(P<0.01,P<0.05)。结论灯盏生脉胶囊可降低氯吡格雷的最大血药浓度和延长体内平均驻留时间,提示临床两药联用时,要注意调整用药剂量与服药间隔时间。Objective To investigate the effect of Dengzhan clopidogrel and its main metabolite SR26334 in rats. Methods Shengmai Capsule (DSC) on the pharmacokinetics of SD rats were randomly divided into two groups, and were orally given clopidogrel, and DSC plus clopidogrel respectively. Blood samples were taken at different time points for the simultaneous determination of blood concentrations of clopidogrel and its carboxylic acid metabolite SR26334. Results Compared with clopidogrel group, the combination group had lower blood maximum concentration (Cmax) of clopidogrel and longer mean retention time(MRT0-t), and the differences were statistically significant(P〈 0.01, P 〈 0.05). Conclusion DSC would decrease the maximum blood concentration and prolong the mean retention time of clopidogrel. It is suggested that we should adjust the dosage and the administration interval when using clopidogrel and DSC together for the treatment of ischemic stroke.
