摘 要:目的建立紫外分光光度(UV)法测定白头翁提取物中总皂苷的方法,并对结果进行不确定度评定。方法采用UV法,进行方法学验证并测定总皂苷,对测定结果进行不确定度评定,分别对来自对照品溶液配制、供试品溶液配制、样品重复性、标准曲线拟合及回收率等因素所引入的不确定度进行评估,由此计算合成不确定度,最终给出测量结果。结果白头翁提取物中总皂苷经不确定度评定表明其含量测定结果在95%置信区间下的扩展不确定度为(75.27±5.42)mg·g-1。结论建立的不确定度评定法能为紫外分光光度法测定中药有效成分的不确定分析提供参考。Objective To establish a quantification method for total saponins in Radix Pulsatillae extracts by UV spectrophotometry and to evaluate the uncertainty of the results. Methods Using UV spectrophotometry, we performed the methodology verification and determined the total saponins in Radix Pulsatillae extracts. The uncertainty factors arising from standard solution preparation, sample solution preparation, sample repetitiveness, calibration curve fitting and recovery tests were evaluated. The final measurement results were present after measurement of the component uncertainty. Results The determined content of total saponins in the uncertainty evaluation showed that the expanded uncertainty of total saponins in the 95 % confidence interval was (75.27 ±5.42)mg·g-1 Conclusion The established mathematic model is applicable to the uncertainty analysis of UV spectrometry in the determination of active components in traditional Chinese medicine.
