摘　　要：目的建立一种快速液相串联质谱测定兔眼房水中原阿片碱含量的方法。方法采用Agilent Eclipse XDB C18（100mm×2.1mm,1.8μm）色谱柱,柱温：35℃,流动相为（A）甲醇-（B）0.1%甲酸溶液梯度洗脱,流速为0.4m L·min^-1。原阿片碱及苦参碱（内标）采用多反应监测正离子模式,原阿片碱定量离子对为m/z 354.2-m/z 188.0,苦参碱定量离子对为m/z 249.1-m/z 148.3。结果原阿片碱在0.5-200ng·m L^-1范围内呈现良好线性关系（r=0.995）,检出限为0.2ng·m L^-1,加样回收率在83.6%-93.7%之间。结论该方法简便、准确,可以用于兔眼房水中原阿片碱含量测定。OBJECTIVE To establish a RRLC-MS / MS method for determination of protopine in rabbit aqueous humor. METHODS The chromatographic separation was performed on an Agilent Eclipse XDB C18（ 100 mm ×2. 1mm,1. 8μm）,the column temperature was set at 35℃. The mobile phase consisting of（ A） methanol-（ B） 0. 1%formic acid solution with the gradient elution was set at a flow rate of 0. 4m L·min%- 1. Protopine and matrine（ internal standard） were detected at positive MRM mode. The MRM ion pairs were： m / z 354. 2-m / z 188. 0 for protopine,m / z249. 1-m / z 148. 3 for matrine. RESULTS A good linear relationship of protopine was showed in 2. 5 - 250 ng ·m L%- 1（ r = 0. 995）,The limit of detection of protopine in rabbit aqueous humor was 0. 2ng·m L%- 1. The recoveries of protopine was in the range of 83. 6% ~ 93. 7%. CONCLUSION The method is accurate and simple,and it can be applied to the determination of protopine in rabbit aqueous humor.
