军丹结肠溶胶囊治疗慢性肾功能不全的药效学研究

摘　　要：目的:评价军丹结肠溶胶囊治疗大鼠肾功能不全的药效。方法:健康成年大鼠80只,进行左肾2/3切除,右肾全切造模,选取存活的60只大鼠,随机分为:模型组、灌胃高剂量组、灌胃低剂量组、灌肠组、空白对照组,每组12只。通过血清肌酐、尿素氮、24 h蛋白尿的检测评价军丹结肠溶胶囊对肾功能的保护作用;通过组织病理学HE染色、PAS染色、Masson染色评价大鼠肾脏的病理改变,探讨军丹结肠溶胶囊可能的作用机制。结果:第12周模型组、中药灌胃高剂量组、中药灌胃低剂量组、中药灌肠组大鼠血肌酐、尿素氮、24 h尿蛋白定量与模型组差别有统计学意义(P<0.01),组织病理改变较模型组减轻。结论:研究表明结肠给药能够延缓慢性肾功能大鼠的肾衰进程,改善剩余肾单位的炎细胞浸润和纤维化,从而起到延缓肾功能下降作用。Objective ： To evaluate the efficacy of Jundan Colon Dissolving Capsule in treatment of renal insufficiency in rats. Methods： 80 adult rats were given left kidney 2/3 resection and total resection of fight kidney to establish the model ,60 rats survived ,which were randomly divided into five groups：model group, gastric perfusion high and low dose groups,retention enema group,blank control group, 12 rats in each group. Evaluate the protective effect of Jundan Colon Dissolving Capsule on renal functions by detecting serum creatinine, urea nitrogen, 24 h urinary protein and observe the pathological change by histopathology HE staining, PAS staining and Masson staining to study the mechanism of Jundan Colon Dissolving Capsule. Results ： The serum creatinine, urea nitrogen and 24 h urinary protein of model group, high and low dose groups of Chinese medicine and herbal enema group had significant difference （ P 〈 0.01 ）. The pathological change was slighter than that of model group. Conclusion： The study showed that colonic delivery can slow down the pro- gression of chronic renal function in rats with renal failure, improve the residual nephron infiltration of inflammatory cells and fibrosis, and thus delaying the decline in renal function effect.
