摘 要:目的:观察消癌解毒方对Walker256大鼠T淋巴细胞亚群和自然杀伤细胞(natural kill,NK)的影响。方法:建立Walker256大鼠模型,50只雄性SD大鼠分为正常组,模型组,中药低剂量组,中药中剂量组,中药高剂量组,每组10只。造模后中药各剂量组进行中药干预,正常组,模型组未进行药物干预。在造模后第10天用流式细胞仪分别检测各组大鼠胸腺,骨髓,外周血T淋巴细胞亚群及NK细胞活性。结果:消癌解毒方干预后,可见低剂量、中剂量组大鼠胸腺指数明显上升,中剂量组大鼠脾指数明显上升,与模型组比较,差异均有显著性(P<0.05);中剂量、高剂量组大鼠骨髓、胸腺细胞与模型组相比,G0/G1期比例显著降低(P<0.05),S期比例明显升高(P<0.05);中剂量组CD4+/CD8+比值明显上升,与模型组相比,差异有显著性(P<0.05);各剂量组NK活性明显增高(P<0.01)。结论:消癌解毒方对Walker256大鼠胸腺指数、脾指数及骨髓、胸腺细胞周期有一定的调节作用,对淋巴细胞亚群CD4+、CD8+、CD4+/CD8+比值及NK细胞活性有一定的调节作用。消癌解毒方具有免疫保护作用可能是其抗癌作用机制之一。Objective:To study the effect of Xiao'ai Jiedu Prescription (XJP)on T lymphocyte subsets and natural killer cells in Walker256 rat model. Methods : Establish Walker256 rat model. 50 male SD rats were divided into normal group, model group, low - dose group, middle - dose group and high - dose group. Treatment group was fed a dose - related XJP. Each group's weight ,thymus index and spleen index were observed. Groups'bone marrow and thymus cells and T cell subsets were detected by flow cytometry. NK kit detected NK cells. Results : It showed that XJP low - dose, middle - dose groups'thymus indexes were significantly increased. The middle -dose group's spleen index was significantly increased in rats. Compared with the model group, the differences were statistically significant ( P 〈 0.05 ). Compared to the model group ,the high- dose group rat bone marrow and thymus ceils GO/GI phase were significantly decreased (P 〈 0.05). Sphase was significantly increased ( P 〈 0.05 ). Compared to the model group, the middle - dose group of CD4+/CD8+ was significantly increased ( P 〈 0.05 ). NK activity of each dose group was significantly increased ( P 〈 0.01 ). Conclusion:XJP can regulate Walker256 rat thymus index, spleen index and bone marrow and thymus cell cycle. XJP can regulate T lymphocyte subsets CD4+ , CD8+ , CD4 +/CD8+ ratio and NK cell activity. XJP with immune protective effect may be one of the mechanisms of its anti - tumor effects.
