调脂积对间断强刺激大鼠血管内皮因子及主动脉超微结构影响的研究

摘  要:目的:通过观察间断强应激刺激干预后老年大鼠模型的血管活性物质一氧化氮、内皮素-1(NO、ET-1)及主动脉超微结构的变化,了解间断强应激刺激对心血管疾病的可能影响;同时观察调脂积对间断强应激刺激后大鼠模型的干预作用。方法:Wistar老年大鼠共32只,随机分为空白组、慢性持续性温和刺激模型对照组、间断强刺激模型组、间断强刺激+调脂积干预组。适应性喂养后,给予2模型组及药物干预组应激刺激(包括冰水游泳、热刺激、禁食、禁水、夹尾、昼夜颠倒、高速水平摇晃、频闪灯照射,但程度及频次不同),间断强刺激+调脂积冲剂干预组在间断强刺激的基础上喂服调脂积冲剂。之后观察各组大鼠体质量变化、糖水偏嗜等一般情况,同时检测血管活性物质(NO、ET-1)水平,电镜下观察主动脉超微结构。结果:给予不同应激刺激56 d后,2模型组与空白组比较,糖水偏嗜度均有降低趋势,但无明显差异(P>0.05);间断强刺激+调脂积干预组与间断强刺激模型组比较,糖水偏嗜度有明显升高,差异明显(P<0.05)。模型组与空白组比较,NO水平明显升高(P<0.05);间断强刺激+调脂积干预组与间断强刺激模型组比较,NO水平有下降趋势,差异显著(P<0.05)。模型组与正常组比较,ET-1水平明显升高(P<0.05);间断强刺激+调脂积干预组与间断强刺激模型组比较,ET-1水平呈下降趋势,但差异不明显(P>0.05)。结论:间断强刺激与慢性持续性温和刺激均可引起大鼠模型相关心血管活性物质(NO、ET-1)及主动脉超微结构变化,可能以此途径影响心血管疾病的发病及预后;调脂积冲剂对间断强刺激引起老年大鼠的相关血管功能及结构的改变有干预作用。Objective : By observing the intermittent strong stress intervention after senile rats models of vascular active substances nitric oxide, endothelin - 1 ( NO and ET - 1 ) and the changes of the ultrastructure of the aorta to understand the intermittent strong stress may affect cardiovascular disease and observe Tiaozhiji intervention on intermittent strong stress rats. Methods :32 elderly Wistar rats were randomly divided into blank group and chronic persistent mild stimulation model control group, intermittent strong stimulation model group, intermittent strong stimulation + Tiaozhiji intervention group. After adaptive feeding,2 model groups and drug intervention groups were given the stress (including ice -water swimming,thermal stimulation,fasting, banning water, tail clip, day and night upside down, high- speed horizontal sha- king,flashing light illumination, but different degree and frequency). The intermittent strong stimulation + Tiaozhiji group on the basis of intermittent strong stimulation was given Tiaozhi Granule. Observe rats weight changes, partial ad- dicted to sugar water,etc. Generally,at the same time detect vascular active substances (NO and ET - 1 ) levels and aor- tic ultrastructure was observed under electron microscope. Results:Given different stress after 56 days, compare blank group and 2 model groups, partial addicted to sugar water degrees reduced with no obvious difference ( P 〉 0.05 ). When intermittent strong stimulation + Tiaozhiji group compared with intermittent strong stimulation model group, the sugar wa- ter partial heavy degree obviously increased ( P 〈 0.05 ). When 2 model groups compared with blank group, the NO level increased significantly ( P 〈 0.05 ). When intermittent strong stimulation + Tiaozhiji group compared with intermittent strong stimulation model group, the NO level had a downturn with significant difference (P 〈 0.05).
