摘  要:目的通过体外研究探索中药淫羊藿苷治疗关节置换假体周围骨溶解的有效浓度。方法将无菌性钛颗粒、含有不同浓度的淫羊藿苷培养液(低浓度、中浓度、高浓度)、小鼠成骨细胞放在一起共同培养。每个浓度对应的用药时间分别设定为6 h、12 h、24 h、48 h,从成骨细胞提取总RNA和mRNA后,采用实时荧光定量PCR方法检测成骨细胞的RANKL-RANK OPG系统的表达变化。结果 RANKL:淫羊藿苷低中高浓度组表达均不同程度下降,中浓度组更显著;OPG:各组均提高了mRNA的表达,中浓度组较明显;OPG/RANKL比率:高浓度组比率增大较明显。结论低中高浓度的淫羊藿苷对钛颗粒引起的成骨细胞的骨溶解作用均有抑制作用,且中高浓度明显。Objective To explore the effective concentration of epimedium glycoside on treating periprosthetic osteolysis after joint replacement by extracorporeal studies. Methods The sterility of titanium particles,epimedium glycosides containing different concentrations of broth( with low density,medium density and high density) and mouse osteoblasts were cultured together. The total RNA and mRNA was got from osteoblasts after each drug concentration corresponding time was set to 6 h,12 h,24 h,48 h. The expression of osteoblast RANKL-RANK OPG system was detected by real-time quantitative PCR method. Results RANKL: Expression in three concentration groups was decreased,in which the decrease of medium density was mostly significant. OPG: The expression of mRNA in three groups was improved,obviously in the middle group. OPG / RANKL ratio: The high concentration group rate increased apparently.Conclusion Various concentrations of epimedium glycoside can inhibit titanium particle-induced osteolysis osteoblasts,aparently in medium and high concentrations.
