肥胖对哮喘鼠模型NF-κB表达影响的研究

摘　　要：目的探讨肥胖和哮喘之间的关联,以及NF-κB在哮喘气道重塑中的作用。方法选20只雄性ob/ob鼠,经过8周肥胖鼠造模成功后,随机分成肥胖组和肥胖哮喘组,再选10只雄性豚鼠为对照组。然后用卵蛋白(ovalbumin,OVA)溶液(1 m L生理盐水+100 mg氢氧化铝+100 mg OVA)制备哮喘组模型,对照组与肥胖组将OVA液用生理盐水代替,成功后取右肺中叶部分,用HE染色和免疫组化进行肺组织切片观察和检测肺组织NF-κB的表达。结果肺组织病理变化:对照组无明显炎症改变,肥胖组肺组织标本呈轻度炎症变化,肥胖哮喘组肺组织标本呈显著炎症变化;免疫组化检测肺组织NF-κB的表达:对照组阳性表达细胞少量,肥胖哮喘组的阳性表达明显高于肥胖组。结论肥胖为哮喘的危险因素,可增加哮喘发生频率及严重程度;NF-κB可能在哮喘发生、发展中起到了至关重要的作用。Objective To investigate the association between obesity and asthma,as well as the role of NF-κB in asthmatic airway remodeling. Methods Twenty male ob / ob mice which were induced into obese rat models successfully in eight weeks were randomly divided into obesity / asthma rat group and obesity group,10 rats in each group,and another 10 male guinea pigs were as control. The rats in asthma group were treated with OVA fluid（ 1 m L normal saline solution ＋ 100 mg aluminum hydroxide ＋ 100 mg OVA）,and rats in obesity group and control group were treated with normal saline solution. After successful induction,the middle of the right lung sections were taken from these rats and the lung biopsy were studied with HE staining and immunohistochemical assay was used to observe and measure the NF-k B expression in the lung tissues. Results There was no significant inflammatory changes in the control group,while lung specimen showed mild inflammatory changes in the obesity group,and significantly increased volume of inflammatory changes in obesity / asthma group; A few positive cells of NF-κB expression were found in control group by immunohistochemistry,while NF-κB level in obesity / asthma group was significantly higher than that in obesity group. Conclusion Obesity is an independent risk factor for the development of asthma,it may increase the frequency and severity of asthma; NF-κB may play a vital role in the development of asthma.
