摘  要:目的建立大鼠动脉粥样硬化模型,观察血液生化指标的变化.方法选取20只健康成年雄性SD大鼠,随机分为2组:正常对照组和模型组,每组10只.正常对照组大鼠给予基础饲料喂养,模型组大鼠在腹腔注射VD3基础上,分阶段先后给予2种不同的高脂饲料喂养.喂养20周后腹主动脉采血,测定血液生化指标,包括血脂(TC、TG、LDL-C、HDL-C)、血糖、肝功指标(ALT和AST),取胸主动脉进行常规组织病理学检查.结果高脂饲料喂养20周可成功建立大鼠动脉粥样硬化模型.模型组大鼠TC和TG水平明显升高,与正常对照组比较,差异具有统计学意义(P<0.05).模型组大鼠血糖水平略高于正常对照组,但差异无统计学意义(P>0.05).模型组大鼠血浆ALT和AST水平分别明显高于常对照组,差异有统计学意义(P<0.05).结论高脂饲料长期喂养建立大鼠动脉粥样硬化模型过程中,可引起大鼠高脂血症和一定程度的肝功能损伤.Objective To establish a rat model of atherosclerosis, and observe the changes of blood biochemical indexes of rats. Methods 20 SD rats (male) were randomly divided into two groups: normal control group and model group (n = 10 in each group) . Rats in the normal control group were only given the basal diet. Rats in the model group were given sequential feeding two kinds of high-fat diets on the basis of intraperitoneal injection of VD3 (750 000 U/kg) . After feeding for 20 weeks, blood samples were collected from the abdominal aorta of rats in two groups to detect the blood biochemical indexes, including blood lipids (TC, TG, LDL-C and HDL-C) , blood glucose and liver function indexes (ALT and AST) Thoracic aorta was collected for histopathological examination. Results Sequential feeding two kinds of high-fat diets for 20 weeks successfully established a rat model of atherosclerosis. The plasma lipids (TC and TG) and plasma ALT and AST levels of rats were significantly higher in the model group than those in the normal control group, the difference had statistical significance (P 〈 0.05) . The blood glucose levels of rats in the model group were a little higher than those in the normal control group, but the difference had no statistical significance (P 〉 0.05) . Conclusion Long-term feeding high-fat diets may lead to hyperlipoidemia and liver function impairment to some extent in establishing rat model of atherosclerosis.
