摘 要:目的研究山竹壳提取物对小鼠成骨样细胞株MC3T3-E1增殖和分化的影响。方法采用活性追踪的方法,用不同终质量浓度的山竹壳提取物和D101大孔树脂柱层析样品溶液进行干预,分别以MTT法和细胞内碱性磷酸酶(ALP)活性检测法评价各样品对MC3T3-E1细胞增殖和分化的影响;用试管反应方法鉴别活性部位的主要化学成分类型,再按照2010版《中国药典》附录XB法测定活性部位的鞣质含量。结果山竹壳水溶性成分有一定的促MC3T3-E1细胞增殖和分化的作用,而脂溶性成分具有显著的细胞毒性;水溶性成分经D101大孔树脂柱层析的70%乙醇洗脱部位(SZ-S7)在12.5~50.0 mg/L范围内培养24、48、72 h时均可显著促进MC3T3-E1细胞增殖,培养3、5、7 d时均能显著促进MC3T3-E1细胞内ALP的形成,且其作用明显优于其它洗脱部位;化学成分分析表明,SZ-S7主要为缩合鞣质,总缩合鞣质含量达87.8%。结论山竹壳水溶性成分具有促进小鼠成骨样细胞株MC3T3-E1的增殖和分化的作用,且经D101大孔树脂柱层析的70%乙醇洗脱部位为主要活性部位,可能对预防和治疗骨质疏松有一定的作用。Objective To investigate the effects of extracts of pericarp of Garcinia mangostana on proliferation and differentiation of mouse osteoblast-like cells MC3T3-E1. Methods Bioactivity-guided separation was carried out on the 80% Et OH extract of the pericarp of Garcinia mangostana. The aqueous-soluble part was chromatographied by D101 macroporous resin column. MTT method was adopted in proliferation analysis and the intracellular alkaline phosphatase(ALP) activity was assayed in differentiation detection after incubated with different doses of extractions and fractions for different times. The chemical constituent of the active fraction was identified by tube tests and its tannin content was measured according to Appendix XB of Pharmacopoeia of People's Republic of China(2010 Edition Subdivision One). Results The aqueous-soluble part could increase proliferation and differentiation of MC3T3-E1 cells, while the ethyl acetate-soluble part had significant cytotoxicity to MC3T3-E1 cells. The 70% Et OH eluted part(SZ-S7) from D101 column at doses ranging from 12.5 mg/L to 50.0 mg/L could remarkably promote the proliferation of MC3T3-E1 after incubated 24, 48 and 72 h, and the ALP activity after incubated 3, 5 and 7 days, respectively, which were better than other eluted parts. Based on the chemical analysis, the major constituent of SZ-S7 was detected as condensed tannin, whose content was up to 87.8%. Conclusion The aqueous-soluble part of the pericarp of Garcinia mangostana has effects on stimulating the proliferation and differentiation of MC3T3-E1 cells, and its active constituent is recognized as condensed tannin in the 70% Et OH eluted part(SZ-S7) from D101 column, which supposes that the pericarp of Garcinia mangostana may have potential in preventing osteoporosis, and deserve further study.
