半夏泻心汤对胃腺癌SGC-7901细胞形态及STAT3通路相关蛋白的影响

摘　　要：目的:研究半夏泻心汤及其拆方对胃腺癌SGC-7901形态的影响及诱导其凋亡的机制。方法:把半夏泻心汤拆分为苦降(黄芩:黄连=3:1)、辛开(干姜:半夏=3:4)、补益(人参:甘草=3:3)3个组方,采用倒置显微镜观察用药后细胞形态变化,采用S-P免疫组化法检测半夏泻心汤全方及其拆方组对凋亡相关基因STAT3、P-STAT3的表达。结果:倒置显微镜下显示,不同浓度的半夏泻心汤全方组、苦降组(黄芩、黄连)及辛开组(干姜、半夏)作用于SGC-7901胃癌细胞株48小时后均能使细胞密度下降,细胞出现变圆、萎缩、不贴壁的现象;高浓度下的半夏泻心汤全方组,苦降组(黄芩、黄连),辛开组(干姜、半夏)作用于SGC-7901胃癌细胞48h后均能使STAT3、P-STAT3蛋白表达降低,与空白组阳性比例(39.12%,41.12%)相比,均有明显降低(14.45%、13.65%,13.41%、17.32%,21.42%、19.01%),其效果强于低浓度,补益组(人参、甘草)则无效。结论:半夏泻心汤全方及苦降组(黄芩、黄连)、辛开组(干姜、半夏)可抑制肿瘤细胞的生长,改变细胞的形态。其抑制细胞生长可能与干预STAT3转录因子的表达,促进肿瘤细胞凋亡有关。Objective: To study the influence of morphology of gastric adenocarcinoma SGC7901 tumor cell and its apoptosis mechanism by Banxia Xiexin decoction and its decomposed recipes. Methods: Banxia Xiexin decoction was divided into bitter descending( coptis chinensis,scutellaria) group,pungent opening( dried ginger,pinellia ternate) group,and invigoration( glycyrrhiza,ginseng) group by its different nature and flavor. The morphological changes were observed before and after treatment by the inverted microscope. The expression of apoptosis associated gene STAT3,P-STAT3 of Banxia Xiexin decoction and its decomposed recipes in SGC7901 tumor cell line were detected by S-P( Phosphate-Peroxidase) immunohistochemical method. Results: Under the inverted microscope,the different concentrations of Banxia Xiexin whole recipe group,bitter descending( coptis chinensis,scutellaria) group,and pungent opening( dried ginger,pinellia ternate) can reduce the density of cell,the cells became round,atrophy,and not adherence. The high concentration of Banxia Xiexin whole recipe group,bitter descending( coptis chinensis,scutellaria) group,and pungent opening( dried ginger,pinellia ternate) can decrease the STAT3 protein expression and the P-STAT3 protein expression with affecting gastric adenocarcinoma SGC7901 tumor cell after 48 hours,compared with the positive proportion of blank group( 39. 12%,41. 12%),it was significantly reduced( 14. 45%,13. 65%,13. 41%,17. 32%,21. 42%,19.01%),which was better than low concentration,while invigoration group( glycyrrhiza,ginseng) had no such effects. Conclusion: Banxia Xiexin decoction and bitter descending( coptis chinensis,scutellaria) group,and pungent opening( dried ginger,pinellia ternate) group can inhibit the growth of tumor cells,change the morphology of the cells; which may related to intervening the expression of transcription factor STAT3,and promote tumor cell apoptosis.
