摘 要:目的:观察雷公藤多苷对糖尿病肾病大鼠白细胞介素1β(IL-1β)、白细胞介素17(IL-17)、干扰素γ(IFN-γ)和单核细胞趋化蛋白-1(MCP-1)等炎性细胞因子表达的影响,讨论其对糖尿病肾病大鼠肾脏炎症损伤的改善作用,分析其可能的治疗机制。方法:将85只雄性SD大鼠分为正常对照组(n=13)和模型组(n=72),采用腹腔注射链脲佐菌素(STZ)的方法建立糖尿病肾病大鼠模型,其中70只大鼠造模成功。将其随机分为模型组、雷公藤多苷治疗组(4.5 mg/kg、9 mg/kg和18 mg/kg)以及罗格列酮(0.4mg/kg)组。末次给药后,检测大鼠血糖(BG)、血甘油三酯(TG)、血总胆固醇(TC)、尿素氮(BUN)、血肌酐(Scr)和24 h尿蛋白量(24 h UAER);放射免疫分析法测定血清中IL-1β水平,酶联免疫吸附法测定各组血清IL-17、IFN-γ的水平;免疫组织化学染色检测肾组织中MCP-1的表达;HE染色观察大鼠肾脏病理变化。结果:与模型组相比,雷公藤多苷9 mg/kg组和18 mg/kg组大鼠的一般情况明显好转,BG、TG、TC、BUN、Scr、24 h UAER的水平显著下降;雷公藤多苷18 mg/kg组大鼠的IL-1β、IL-17、IFN-γ和肾组织中MCP-1的表达明显下调,肾脏病理变化明显减轻。结论:雷公藤多苷可能是通过降低血清及肾脏炎性因子的水平,延缓肾脏的损伤,从而对肾脏起到一定程度的保护作用。Objective :To observe the protective effect of tripterygium wilfordii polyglycoside (TWP) on the expression of interleukin-113 (IL-113), interleukin-17( IL-17), Interferon-3,( IFN-3,) and monocyte chemotactic protein-1 (MCP-1) in diabetic nephropathy rats, and to investigate the protective effect on inflammatory injury renal. Methods: 85 male SD rats were randomly divided into normal control group ( n = 13 ) anddiabetic nephropathy(DN) model group (n = 72). The DN model was induced by intraperitoneal injection of streptozotocin(STZ). 70 DN model rats were randomly divided into model control group, drug TWP groups(4. 5 mg/kg,9 mg/kg and 18 mg/kg) and rosiglitazone (0.4 mg/kg) group. After the last administration, to detect blood glucose( BG), triglycerides (TG), total cholesterol (TC), blood urea nitrogen ( BUN), serum creatinine ( Scr), and 24 hours urinary albumin excretion rate(UAER) ; The expression of the serum IL-17, IFN-α were ex- amined by ELISA ; IL-1 β were determined by radioimmunoassav. The pathological changes of kidney were observed by HE staining and the expression of MCP-1 in kidney was tested by immunohistochemistry. Results: Compared with the model group, the general status of the rats in TWP (9 mg/kg and 18 mg/kg) were significantly improved; levels of BG, TG, TC, BUN, Scr, 24 h UAER were signiLificantly decreased (P 〈0.01 ); The expression of IL-1β, IL-17, IFN-y and MCP-1 in kidney of TWP (18 mg/kg) decreased obviously, renal pathological changes were significantly alleviated. Conclusion: TWP could protect the renal tissue by decreasing the inflammatory factors, and inhibiting pathological changes.
